摘 要:目的观察不同浓度复方夏枯草制剂对人非小细胞肺癌细胞(NCI-H460)移植裸鼠瘤体生长的影响,并探讨其可能机制。方法取SPF级裸鼠36只,将0.2 mL的NCI-H460细胞悬液注射到裸鼠右后肢皮下制备移植瘤模型,将肿瘤体积长到60~80 mm3的30只裸鼠(其余6只剔除实验)分为高剂量组、中剂量组、低剂量组、阳性组、模型组各6只,另取6只正常裸鼠作为对照组,高剂量组、中剂量组、低剂量组分别以2.4、1.2、0.6 g/kg的复方夏枯草制剂灌胃,阳性组注射0.025 g/kg的环磷酰胺,对照组和模型组灌胃等体积蒸馏水;阳性组隔两天给药1次,其他组每天1次。停药次日,测量移植瘤体积,观察瘤组织病理变化;免疫组化法检测瘤组织Bcl-2、Bax、Caspase-3。结果与模型组比较,高剂量组、阳性组给药后瘤体积、瘤重减小(P均<0.05)。模型组癌细胞紧密,核仁体积大;阳性组和复方夏枯草制剂低、中、高剂量组细胞有不同程度坏死,癌细胞数目明显减少,且大小不一,胞质破裂,完整性不好。与模型组比较,高剂量组、中剂量组、低剂量组、阳性组Bax阳性率升高,高剂量组、中剂量组、阳性组Caspase-3阳性率升高(P均<0.05)。结论复方夏枯草制剂对NCI-H460细胞移植裸鼠瘤体生长有抑制作用,高剂量浓度效果最好,其作用机制可能与上调Bax、Caspase-3表达有关。Objective To observe the effects of different concentrations of compound Prunella vulgaris preparation on tumor growth of human non-small-cell lung cancer(NSCLC)NCI-H460 cells-transplanted nude mice.Methods We selected 36 SPF-class nude mice and injected 0.2 mL NCI-H460 cell suspension into the right hind limb of nude mice to prepare the transplanted tumor models.Thirty nude mice whose tumor volume was 60-80 mm3 were randomly divided into five groups(n=6):model group,positive control group,low-dose,medium-dose and high-dose groups,and another 6 normal nude mice were used as the normal control group.The mice in the high-dose,medium-dose and low-dose groups were orally administered with compound Prunella vulgaris preparation at the dosage of 2.4,1.2,and 0.6 g/kg,respectively;the mice in the model group and normal control group were orally administered with equal volume of distilled water;and the mice in the positive control group were injected with 0.025 g/kg cyclophosphamide(CTX).Except the positive control group of once every two days,the other groups were given once a day.The second day after withdrawal,the volume of the transplanted tumor was measured,and the pathological changes of the tumor tissues were observed.The Bcl-2,Bax,and Caspase-3 in the tumor tissues were detected by immunohistochemistry.Results Compared with the model group,the tumor weight and volume of the positive control group and high-dose group decreased significantly(P<0.05).In the model group,the cancer cells were tight and the nucleolus was large.Moreover,the cells of the positive control group,low-dose group,medium-dose group,and high-dose group had different degrees of necrosis,and the number of cancer cells decreased,and with different size,meanwhile,the cytoplasm was broken and the integrity was not good.Compared with the model group,the Bax positive expression rates in the tumor tissues of the low-dose,medium-dose and high-dose groups,and positive control group increased(all P<0.05),and the Caspase-3 positive expression rates in the m
