摘  要:目的:探讨逍遥解郁方对卒中后抑郁大鼠5-羟色胺(5-HT)及其受体表达水平的影响和作用机制。方法:SD大鼠50只,随机分为空白组、模型组、氟西汀组、中药低、高剂量组,每组10只。首先采用大脑中动脉结扎(MCAO)术建立卒中模型,在此基础上采用慢性温和不可预知应激(CMUS)结合孤养建立抑郁模型。卒中后抑郁模型建立后,药物连续灌胃28 d后检测各组大鼠血清中5-HT及其脑组织中受体表达。结果:与空白组比较,各组大鼠血清5-HT含量明显降低(P<0.01)。与模型组比较,各组大鼠血清5-HT浓度明显增加伴海马5-HT_(1A)R表达增加(P<0.05),中药低剂量组5-HT_(2A)R表达无统计学差异(P>0.05),但中药高剂量组和氟西汀组5-HT_(2A)R表达明显减少(P<0.05);与中药低剂量组比较,中药高剂量组和氟西汀组5-HT_(2A)R表达亦明显减少(P<0.01),且两组间比较有统计学差异。结论:逍遥解郁方可显著增加PSD大鼠5-HT含量,其作用机制可能是通过上调5-HT_(1A)RmRNA,下调5-HT_(2A)R mRNA表达来实现的。Objective:To investigate the effect of Xiaoyao Jieyu recipe on the levels of 5-hydroxytryptamine(5-HT) and its receptors in the rats with post-stroke depression(PSD)and its potential mechanisms.Methods:50 SD rats were selected and randomly divided into blank group,model group,fluoxetine group,low-dose-traditional Chinese medicine(TCM) group and high-dose-TCM group,with 10 rats in each group.The stroke model was established by middle cerebral artery occlusion (MCAO),and then the depression model was established by combining chronic unpredictable mild stress (CUMS) with solitary nursing.As the post-stroke depression model established,serum 5-HT concentration and its receptor expression in the brain tissues of rats in each group were detected after 28 days of continuous gavage.Results:Compared with the blank group,serum 5-HT concentration was significantly reduced in each group (P<0.01).Compared with the model group,serum 5-HT concentration in each group was significantly increased with an increase in the expression of 5-HT 1A R in the hippocampus (P<0.05).And there was no significant difference in 5-HT 2A R expression in the low-dose-TCM group (P >0.05),while a significant reduce in 5-HT 2A R expression in the high-dose-TCM group and the fluoxetine group (P<0.05).Compared with the low-dose-TCM group,the 5-HT 2A R expression in the high-dose-TCM group and the fluoxetine group was significantly reduced (P<0.01),and there was no significant difference between the two groups (P >0.05).Conclusion:Xiaoyao Jieyu recipe could significantly increase serum 5-HT concentration of PSD rats and its mechanism may be achieved by up-regulating the expression of 5-HT 1A R and down-regulating that of 5-HT 2A R.
