摘 要:Paracoccus sp. QD15-1是一株高效酞酸酯降解菌,在酞酸酯污染修复方面具有较好的利用前,而廉价发酵基质是应用于生产实践的前提。本实验以Paracoccus sp. QD15-1为研究对象,用爬坡法来确碳源、氮源和微量元素的最佳范围,通过响应面中心合成设计法优化发酵培养基。研究结果表明,最优的碳源为玉米面粉,氮源为大豆面粉;经爬坡实验确碳源、氮源和痕量元素分别为10,20,1.5mL/L;响应面实验确最优培养基基质为玉米面粉9.5 g/L、大豆面粉18.6 g/L和痕量元素1.64 mL/L,预测Paracoccus sp. QD15-1的菌落数4.67×109 CFU/mL,实测菌落数为4.5×109 CFU/mL。相较于DMP无机盐培养基菌量提高22.22%,廉价发酵培养基大幅降低生产成本,为大规模发酵生产Paracoccus sp. QD15-1提供重要技术支持。Paracoccus sp.QD15-1 is a high-efficiency phthalate-degrading bacterium,which has a good utilization prospect in the recovery of phthalate pollution,and the cheap fermentation matrix is a prerequisite for production practice.In this experiment,Paracoccus sp.QD15-1 was used as the research object.The climbing slope method was used to determine the optimal range of carbon source,nitrogen source and trace elements.The fermentation medium was optimized by using the response surface center synthesis design method.The results show that the optimal carbon source is corn flour and the nitrogen source is soybean flour.The carbon source,nitrogen source and trace elements are determined by climbing test was 10 g/L,20 g/L and 1.5 mL/L,respectively.the optimized medium composition was:corn flour 9.5 g/L,soybean flour 1.86 g/L and trace elements 1.64 mL/L,the number of colonies of Paracoccus sp.QD15-1 was 4.67×109 cfu/mL.Compared with the DMP inorganic salt medium,it increased by25.05%,which greatly reduced the production cost,and provided important technical support for the large-scale fermentation production of Paracoccus sp.QD15-1.
