摘　　要：目的探讨长链非编码生长停滞特异性蛋白6-反义RNA1(lncRNA DLX6-AS1)在非小细胞肺癌(NSCLC)癌组织中的表达及对NSCLC癌细胞增殖和凋亡的调节作用。方法收集2011-2016年于该院胸外科行手术治疗23例NSCLC患者的癌组织和癌旁组织,培养人支气管上皮细胞(16HBE)、NSCLC细胞系(腺癌:A549、SPC-A1;鳞癌:SK-MES-1),运用实时定量聚合酶链式反应(qRT-PCR)检测DLX6-AS1的表达。运用LipofectamineTM2000转染DLX6-AS1siRNA至A549细胞内干扰DLX6-AS1表达后,四甲基偶氮唑蓝比色(MTT)法检测细胞活力,重组人半胱天冬酶-3(caspase-3)活力试剂盒检测细胞caspase-3活力,Western blot检测凋亡相关蛋白B细胞淋巴瘤-2(Bcl-2)和Bcl-2相关X蛋白(Bax)的表达。结果 lncRNA DLX6-AS1在NSCLC癌组织中表达上调,且与淋巴结转移和临床分期呈明显正相关(P<0.01)。相比16HBE,NSCLC腺癌细胞系A549、SPC-A1和鳞癌细胞系SK-MES-1中lncRNA DLX6-AS1表达均上调(P<0.05)。干扰DLX6-AS1表达可抑制A549的增殖(P<0.05)。lncRNA DLX6-AS1siRNA可增强caspase-3活力,并上调Bax表达和下调Bcl-2表达(P<0.05)。结论lncRNA DLX6-AS1在非小细胞肺癌中的表达上调,促进NSCLC细胞的增殖、抑制凋亡。Objective To investigate the expression of long chain noncoding growth stasis specific protein 6 antisense RNA1(lncRNA DLX6-AS1)in non-small cell lung cancer(NSCLC)and its role on proliferation and apoptosis of NSCLC cells.Methods The cancer tissue and paracancerous tissue in 23 patients with NSCLC treated by thoracic operation in our hospital during 2011-2016 were collected and their human normal bronchial epithelial cells 16HBE,NSCLC cell lines(adenocarcinoma:A549,SPC-A1,squamous cell carcinoma:SK-MES-1)were cultured.The real-time quantitative PCR was used to the expression of lncRNA DLX6-AS1.lncRNA DLX6-AS1 siRNA was transfected into cell by Lipofectamine TM 2000.The cell viability was measured by MTT kit,caspase-3 activity kit was used to evaluated the caspase-3 activity.The expression level of apoptosis related protein B lymphoma 2(Bcl-2)and Bcl-2 related X protein(Bax)were measured by western blot.Results The expression of lncRNA DLX6-AS1 was up-regulated in NSCLC carcinoma tissue,moreover was positively correlated with NSCLC clinical classification and lymph node metastasis(P<0.01).The expression of lncRNA DLX6-AS1 was also up-regulated in NSCLC carcinoma cell lines A549,SPC A1 and squamous carcinoma cell line lncRNA AS1(P<0.05).Interfering DLX6-AS1 expression inhibited the cell proliferation of A549(P<0.05).lncRNA DLX6 AS1 siRNA enhanced caspase-3 activity,upregulated the Bax expression and downregulated Bcl-2 expression(P<0.05)Conclusion The expression of lncRNA DLX6-AS1 is up-regulated in NSCLC,promotes the proliferation and inhibits the apoptosis of NSCLC cells.
