摘　　要：目的观察止血胶对应激性溃疡的治疗作用并探讨其作用机制。方法 30只成年雄性SD大鼠随机分为5组:正常对照组、大平台组、应激组、止血胶组,铝镁加组,每组6只。连续使用小站台水环境法制备应激性溃疡模型。待造模及药物处理结束后,按照Guth标准测定溃疡指数; HE染色法观察胃黏膜损伤; ELISA法检测血清EGF、IL-2、PAF水平;免疫荧光法检测胃黏膜VCL、FAK表达。结果与正常对照组及大平台组相比,单纯应激组大鼠的UI明显增加,EGF、IL-2升高,PAF表达降低,胃黏膜VCL、FAK蛋白表达显著降低,差异均有统计学意义(P <0. 05)。与单纯应激组相比,止血胶组和铝镁加组UI明显降低,血清EGF及IL-2表达显著降低,PAF表达升高,胃黏膜VCL、FAK蛋白表达显著升高,差异均有统计学意义(P <0. 05)。结论止血胶对胃黏膜具有很好的保护作用,其机制可能与血清EGF、IL-2、PAF水平变化及稳定细胞连接相关蛋白VCL和FAK有关。Objective To investigate the therapeutic effect of hemostatic adhesive on stress ulcer and its mechanism.Methods Thirty male SD rats were randomly divided into five groups:control group,big platform group,stress group,hemostatic adhesive group,and almagate group.Animal models of stress ulcer were set up by small platform over water.At the end of the experiment,Guth standard was used to detect ulcer index.HE staining was used to observe gastric mucosa injury.ELASA was used to determine serum levels of EGF,IL-2,PAF.The expression of VCL and FAK in gastric mucosa was detected by means of immunofluorescence.Results Compared with control group and big platform group,the ulcer index(UI),serum levels of EGF,IL-2 were significantly increased,while serum level of PAF and expression of VCL and FAK in gastric mucosa were significantly decreased in stress group(P<0.05).Compared with stress group,the ulcer index(UI),serum levels of EGF,IL-2 were decreased,while serum level of PAF and expression of VCL and FAK in gastric mucosa were markedly elevated in hemostatic adhesive group and almagate group(P<0.05).Conclusion Hemostatic adhesive has good protective effect on gastric mucosa,which may be related to the changes of serum EGF,IL-2,PAF and stabilizing cellular connection-related proteins VCL and FAK.
