清肺养阴方治疗大鼠放射性肺损伤的实验研究

摘　　要：目的探讨清肺养阴方对大鼠放射性肺损伤的治疗作用。方法选取60只雄性Wistar大鼠随机分为空白组、模型组、研究组。用6 MV-X线直线加速器对模型组、研究组大鼠双肺进行单次15 Gy照射,照射后第1天开始灌胃给药(研究组为清肺养阴方,空白组和模型组为蒸馏水),连续灌胃至照射后5周,取大鼠右上肺组织行HE染色观察病理改变;酶联免疫吸附试验(ELISA)检测每周各组大鼠血清中肿瘤坏死因子-α(TNF-α)、转化生长因子-β(TGF-β)和白介素-6(IL-6)的水平。结果 HE染色结果显示:模型组大鼠肺组织第4周放射性肺损伤最显著;研究组大鼠肺组织放射性肺损伤较模型组明显减轻。ELISA结果显示:研究组大鼠血清中TNF-α、TGF-β和IL-6水平显著低于模型组,差异均有统计学意义(P均<0.05)。结论清肺养阴方治疗放射性肺损伤具有较好作用,这可能与其能降低TNF-α、TGF-β和IL-6等细胞因子水平有关。Objective To investigate the effects of qingfei yangyin formula in the treatment of radioactive lung injury in rats.Methods Sixty male Wistar rats were randomly divided into the blank control group,the model group,the treatment group.The whole chest of rats in the model group and the treatment group were irradiated by 6 MV X-rays for 15 Gy dose once.The corresponding drug was given by gastric perfusion once a day from the first day on radiation to 5 weeks after radiation.Right lung was collected,HE stain was used to observe the pathological changes.enzyme linked immunosorbent assay(ELISA)method was used to check the serum levels of tumor necrosis factor-α(TNF-α),transforming growth factor-β(TGF-β)and interleukin-6(IL-6)every week.Results The radioactive lung injury of the model group was the most severe,the radioactive lung injury at the fourth week of the treatment group were better than the model group.Compared with the model group,the expression levels of TNF-α,TGF-βand IL-6 in the treatment group were significantly reduced(P<0.05).Conclusion Qingfei yangyin formula can reduce radioactive lung injury in rats by inhibiting the expressions of TNF-α,TGF-βand IL-6.
