摘  要:目的:局部注射海人酸制作癫痫大鼠模型,研究局部高频电刺激对癫痫大鼠苔藓纤维出芽及痫性发作的影响。方法:海马局部注射海人酸制作大鼠颞叶癫痫模型,选择局部海马癫痫灶进行高频电刺激,选择不同时间点,观察大鼠癫痫发作特点及脑电变化,通过改良Timm染色法,观察海马病理改变。结果:行为和脑电变化,点燃大鼠行为学上可分三期,包括急性期、静止期、慢性期,同时各个时期有相应的脑电特征表现。电刺激可减少慢性期大鼠癫痫发作的次数和痫性放电的次数(P<0.001)。病理改变,点燃后大鼠海马Timm染色可见4 d时苔藓纤维出芽开始出现,到14 d最明显(P<0.01),Golijeh评分电刺激组较海人酸组低(P<0.05)。结论:局部高频电刺激海马能有效抑制海人酸癫痫模型大鼠的癫痫发作及痫性放电,抑制海马苔藓纤维出芽可能是高频电刺激控制癫痫发作的作用机制之一。Objective:In this study,the effects of electrical stimulation on mossy fiber sprouting and epileptic seizure in chronic epileptic rats were investigated by establishing the model of chronic epileptic rats with Kainic acid.Method:The temporal lobe epilepsy models were established with unilateral Kainic acid injections.Local hippocampal epileptic foci were selected for high-frequency electrical stimulation.Different time points were selected to observe the seizure characteristics and EEG changes in rats,the pathological changes of hippocampus was investigated by Timm staining.Result:Behavior and EEG,the behavior of ignited rats can be divided into three phases,including acute phase,quiescent phase and chronic phase,in the three periods,the characteristic of EEG were recorded.HFS may reduce the spontaneous recurrent seizures and epileptic discharge in the chronic phase(P<0.001).Pathology,the Timm staining of the rat hippocampus showed that the mossy fiber sproutings evidence was found at 4 days following induction of SE,and was vigorous at 14 days(P<0.01).IN the Golijeh score,electrical stimulation group was lower than the KA group(P<0.05).Conclusion:High electrical stimulation of hippocampus can effectively inhibit seizures and epileptic discharge in rats with Kainic acid epilepsy.Inhibition of hippocampal mossy fiber sprouting may be one of the mechanisms of high-frequency electrical stimulation to control epileptic seizures.
