摘 要:目的观察电针"百会""大椎""肾俞"对SAMP8小鼠学习记忆与海马神经元NMDA受体NR2A和NR2B表达的影响,探讨电针治疗阿尔茨海默病的机制。方法 6月龄SAMP8小鼠24只,随机分为模型组和电针组,同龄正常老化SAMR1小鼠12只为对照组。电针组电针"百会""大椎""肾俞"30d。Morris水迷宫实验检测小鼠学习记忆能力,免疫组化染色和Western blot检测海马神经元NR2A和NR2B的表达。结果与对照组比较,模型组小鼠逃避潜伏期明显延长(P<0.01),原平台象限停留时间明显缩短(P<0.01),跨越原平台次数明显减少(P<0.01),海马神经元NR2A相对表达量明显升高(P<0.01),NR2B相对表达量明显下降(P<0.01);与模型组比较,电针组小鼠逃避潜伏期明显缩短(P<0.05),原平台象限停留时间明显延长(P<0.01),跨越原平台次数明显增加(P<0.05),海马神经元NR2A相对表达量明显下降(P<0.05),NR2B相对表达量明显升高(P<0.01)。结论电针能改善SAMP8小鼠的学习记忆能力,其作用机制可能与电针抑制海马神经元NMDA受体NR2A的表达和促进NMDA受体NR2B的表达有关。Objective To observe the effects of electroacupuncture (EA) at acupoints of Baihui (GV20), Dazhui (GV14) and Shenshu (BL23) on cognitive function and neurons NMDA receptors NR2A and NR2B in the hippocampus in SAMP8 mice;To explore the mechanism of EA in the treatment of Alzheimer disease (AD). Methods Totally 24 six-month-old SAMP8 mice were randomly divided into model group ( n =12) and EA group ( n =12), and the same age SAMR1 mice were as control group ( n =12). The EA group accepted EA at Baihui, Dazhui and Shenshu for 30 days. Morris water maze was used to detect the learning and memory ability of mice. Immunohistochemical staining and Western blot were used to observe the expressions of NR2A and NR2B in hippocampal neurons. Results Compared with the control group, escape latency increased ( P <0.01), the time staying in the quadrant of the platform decreased ( P <0.01), as well as the number passing the original platform ( P <0.01), and the relative expression of NR2A increased ( P <0.01) and the relative expression of NR2B ( P <0.01) in hippocampus decreased in the model group. Compared with the model group, escape latency decreased ( P <0.05), the time staying in the quadrant of the platform increased ( P <0.01), as well as the number passing the original platform ( P <0.05), and the relative expression of NR2A decreased ( P <0.05) and the relative expression of NR2B increased ( P <0.01) in hippocampus in EA group. Conclusion EA can improve the learning and memory ability of SAMP8 mice, and its mechanism may be that the EA can inhibit the expression of neurons NMDA receptor NR2A and increase the expression of NR2B in hippocampus.
