摘 要:目的姜黄素对甲状腺乳头状癌(PTC)细胞株TPC-1的侵袭和迁移影响研究较少。文中旨在探究姜黄素对PTC TPC-1细胞侵袭和迁移的影响,并探讨可能的机制。方法将TPC-1细胞制成单细胞悬液,分别给予含不同姜黄素浓度的培养基干预分为:0μmol/L组(DMSO溶剂)以及10、20、40、60、80、100μmol/L组。CCK8法检测各组细胞存活率的抑制效应;采用划痕和Transwell实验检测姜黄素对TPC-1细胞迁移和侵袭的影响;采用RT-PCR和Western blot方法检测各组细胞葡萄糖转运蛋白1(Glut1)和膜型基质金属蛋白酶-1(MT1-MMP)mRNA及蛋白的表达水平。结果各组间细胞存活率比较差异均有统计学意义(P<0.05)。24 h后,0、10、20、40μmol/L组细胞迁徙力分别为:[(0.842±0.096)、(0.911±0.049)、(0.926±0.107)、(1.076±0.093)mm],两两组间比较差异有统计学意义(P<0.05);48 h后,组间比较差异亦有统计学意义(P<0.05)。0、10、20、40μmol/L组迁移的细胞数量分别为196、142、57和17个/100倍视野,两两组间比较差异均有统计学意义(P<0.05)。各组间Glut1和MT1-MMP mRNA及蛋白的相对表达量组间比较差异亦有统计学意义(P<0.05)。结论姜黄素具有抑制PTC TPC-1细胞侵袭和迁移能力,可能与降低Glut1和MT1-MMP蛋白表达有关。Objective Comparatively few studies are reported on the invasion and migration of papillary thyroid carcinoma(PTC)TPC-1 cells.This study was to investigate the effects of curcumin on the invasion and migration abilities of TPC-1 cells and its possible action mechanisms.Methods We treated TPC-1 single cell suspension with curcumin at the concentrations of 0(DMSO solvent),10,20,40,60,80 and 100μmol/L.At 24 and 48 hours af-ter exposure,we examined the inhibitory effect of curcumin on the cells by CCK-8 assays,detected the migration and invasion abilities of the TPC-1 cells by Transwell and wound healing assay,and deter-mined the gene and protein expressions Glut1 and MT1-MMP by RT-PCR and Western blot,respectively.Results There were statistically significant differences in the cell viability among different groups of the TPC-1 cells(P<0.05)as well as in the cell migration ability at 24 hours between any two groups of the cells treated with curcumin at 0μmol/L([0.842±0.096]mm),10μmol/L([0.911±0.049]mm),20μmol/L([0.926±0.107]mm)and 40μmol/L([1.076±0.093]mm)(P<0.05)and at 48 hours(P<0.05).Statistically significant differences were also observed between any two of the 0,10,20 and 40μmol/L groups in the number of migrated cells(196,142,57,and 17/100x visual field)(P<0.05)as well in the protein expression of Glut1(0.786±0.112,0.518±0.106,0.359±0.121,and 0.266±0.087)(P<0.05)and the mRNA and protein expressions of MT1-MMP(P<0.05).Conclusion Curcumin can inhibit the invasion and migration of thyroid papillary carcinoma TPC-1 cells,which may be associated with the decreased expressions of Glut1 and MT1-MMP.
