摘  要:目的:探究生物生长促进剂褪黑素对深层发酵灵芝酸含量和产量的影响。方法:通过菌丝体斜面激活、种子液培养、扩大培养三阶段获得灵芝菌丝体,用香草醛-高氯酸法测定其灵芝酸的含量,并计算产量。结果:当培养基中褪黑素浓度为20μmol/L时,灵芝酸含量与灵芝酸产量分别比对照提高了17.61%和46.07%;与其他添加时间相比,第4天时添加20μmol/L褪黑素提高灵芝酸含量与产量最显著;与其他收获菌丝体时间相比,第8天时灵芝酸含量与产量均达到最大值。结论:灵芝菌丝体培养至第4天时加入20μmol/L褪黑素,继续培养至第8天,收获灵芝酸产量最高。Objective:To investigate the concentration and yield of ganoderic acid in submerged fermentation broth thought adding melatonin.Methods:It is through three stages of the mycelia activation,seed culture and submerged fermentation to obtain mycelia of Ganoderma lucidum.The mycelia was ground into powder in order to extract ganoderic acid with ethanol.The concentration of ganoderic acid was determined with vanillic aldehyde-perchloric acid.Results:In the range of experimental concentration,it indicated that the concentration and the yield of ganoderic acid in mycelia treated with 20μmol/L of melatonin were augmented by 17.6%and 46.7%compared with control group,respectively.The concentration and the yield of ganoderic acid in mycelia reached peak when 20μmol/L of melatonin was added to expanded medium on day 4 compared with any other days.The maximum of ganoderic acid yield in mycelia treated with 20μmol/L of melatonin was observed when the mycelia was harvested on day 8.Conclusion:The highest yield of ganoderic acid in mycelia was obtained by adding melatonin of 20μmol/L to the mycelium of Ganoderma lucidum on the 4th day and harvesting on the 8th day.
