摘 要:目的探索骨肉瘤(osteosarcoma,OS)肺转移的外周血清miRNA的表达差异以及与肺转移的相关性。方法选取2017年4月至2018年4月西安交通大学医学院附属红会医院肿瘤外科、西安交通大学第一附属医院肿瘤外科和西安交通大学第二附属医院肿瘤外科的88例OS患者(其中41例发生肺转移)外周血样本,使用miRNA微矩阵芯片预筛选6例患者中与OS肺转移相关的miRNA分子;使用qPCR检测OS肺转移相关的差异miRNA分子的表达;使用单因素Logistic回归分析6个差异miRNA分子与OS肺转移的相关性。结果肺转移与未转移OS患者的血清miRNA差异有统计学意义(P<0.05);Logistic回归分析显示,miR-219-5p、miR-19b-1和miR-25与肺转移关系密切(P<0.05)。结论 OS患者肺转移前后血清miRNA有明显差异;血清miR-219-5p、miR-19b-1和miR-25与肺转移相关性密切,可提示OS肺转移的发生,并作为潜在的预测OS肺转移生物学指标。Objective To explore the expression differences of miRNAs in the peripheral serum of osteosarcoma(OS)with lung metastasis and their correlations with lung metastasis.Methods The peripheral serum samples of 88 OS patients(including 41 cases with lung metastasis)treated in Xi'an Honghui Hospital Affilicated to Xi'an Jiaotong University,The First Affiliated Hospital and The Second Hospital of Xi'an Jiaotong University from April of 2017 to April of 2018 were collected.The miRNA microarray was used to pre-screen the miRNA molecules associated with OS lung metastasis in 6 patients,the qPCR assay was used to detect the expression of the differential miRNA molecules associated with OS lung metastasis,and the single-factor logistic regression analysis was used to analyze the correlations of 6 differential miRNA molecules with OS lung metastasis.Results There was significant difference in the expression of serum miRNA between the patients with OS lung metastasis and those without it.The results of Logistic regression analysis showed that the expression levels of miR-219-5p,miR-19b-1 and miR-25 were closely correlated with lung metastasis(P<0.05).Conclusion The expression levels of serum miRNAs before and after lung metastasis are significantly different in the OS patients and those of serum miR-219-5p,miR-19b-1 and miR-25 are closely correlated with lung metastasis,which can be used as the potential biomarkers for suggesting and predicting the occurrence of OS lung metastasis.
