摘　　要：目的探讨沉默肌球蛋白调节轻链9(MYL9)基因对非小细胞肺癌H1299细胞株增殖及迁移的影响。方法将H1299细胞株分为对照组(CON组)、阴性对照组(NC组)和敲减组(KD组),CON组细胞不进行转染,NC组、KD组分别转染空载短发夹RNA慢病毒和沉默MYL9基因表达的短发夹RNA慢病毒。检测3组细胞MYL9 mRNA表达水平、增殖能力及迁移能力。结果与CON组和NC组比较,KD组MYL9的mRNA相对表达水平、细胞增殖及细胞迁移能力均更低(均P <0. 05)。结论沉默MYL9基因可下调非小细胞肺癌H1299细胞株MYL9基因的mRNA表达水平,降低其增殖和迁移能力。Objective To investigate the effect of silencing myosin light chain 9(MYL9) gene on proliferation and migration of non-small cell lung canceRH1299 cell line.Methods The H1299 cell line was divided into control group(CON group),negative control group(NC group),oRknockdown group(KD group).The cells in the CON group were not transfected,and the NC group and KD group were transfected with empty short hairpin RNA lentivirus and MYL9 gene-silenced short hairpin RNA lentivirus,respectively.The expression levels of MYL9 mRNA,cell proliferation and migration ability of the three groups were detected.Results Relative expression level of MYL9,cell proliferation and migration ability were loweRin KD group than in CON group and NC group(all P <0.05).Conclusion Silencing MYL9 gene can down-regulate the mRNA expression level of MYL9 gene,and reduce the proliferation and migration ability in H1299 cell line of non-small cell lung cancer.
