摘 要:目的制备3Y-TZP陶瓷表面镧(La)掺杂TiO_2抗菌薄膜观察其抗菌性,为临床抗菌氧化锆陶瓷的应用提供实验基础。方法制备直径20 mm、高3 mm圆柱形3Y-TZP试件,采用溶胶凝胶法制备质量分数1.0%镧(La)掺杂的TiO_2溶胶,并通过浸渍提拉法涂覆于3Y-TZP表面制成La-TiO_2薄膜-3Y-TZP陶瓷,依同法,不添加硝酸镧溶液制得TiO_2薄膜-3Y-TZP陶瓷。扫描电镜对3Y-TZP陶瓷试件(3Y-TZP组)、TiO_2薄膜-3Y-TZP陶瓷试件(TiO_2薄膜组)、La-TiO_2薄膜-3Y-TZP陶瓷试件(La-TiO_2薄膜组)进行表面形貌观察,并进行光催化实验、抗菌实验和细胞毒性实验。(1)光催化实验,La-TiO_2薄膜组和TiO_2薄膜组,各随机取4个试件,观察各组试件日光下不同时间点对亚甲基蓝的降解率;(2)抗菌实验,La-TiO_2薄膜组、TiO_2薄膜组和3Y-TZP组,每组随机取5个试件,通过抑菌圈法检测其抗菌性,比较各组抑菌距离;(3)细胞毒性实验,La-TiO_2薄膜组、TiO_2薄膜组、3Y-TZP组(为阴性对照组)每组随机取3个试件,制得浸提液,以0.064%苯酚溶液为阳性对照组。4组培养L-929细胞3 d后进行形态学观察,采用MTT法测试各组细胞活力、细胞相对增殖率。结果采用浸渍提拉法可在3Y-TZP表面制备均匀分布的La-TiO_2薄膜。光催化实验显示La-TiO_2薄膜组在日光下2 h亚甲基蓝降解率达到(41.2±1.5)%,高于TiO_2薄膜组(36.5±2.4)%,两组差异具有统计学意义(t=3.321,P=0.016)。抗菌实验显示,La-TiO_2薄膜组抑菌距离为(0.34±0.08)mm,大于TiO_2薄膜组的(0.12±0.02)mm,2组差异没有统计学意义(F=63.798,P<0.001),3Y-TZP组无明显抑菌圈。细胞毒性实验显示La-TiO_2薄膜组细胞相对增殖率为(89.5±1.3)%,细胞毒性评级为1级;4组间差异具有统计学意义(F=68.250,P<0.001)。结论镧掺杂TiO_2能改善其光催化活性,增强3Y-TZP陶瓷的抗菌性,且无明显细胞毒性。Objective To prepare the La-doped TiO2 film on the surface of 3 Y-TZP ceramics in order to observe its antibacterial properties,providing an experimental basis for the application of antibacterial zirconia ceramics in the clinic.Method A cylindrical 3 Y-TZP specimen with a diameter of 20mm and a height of 3mm was prepared.The solgel method was used to prepare 1%lanthanum(La)-doped TiO2 sol,and the La-TiO2 film-3 Y-TZP ceramic was prepared by dip coating on the surface of 3 Y-TZP.The TiO2 thin film-3 Y-TZP ceramic was prepared by adding no lanthanum nitrate solution in the same way.The surface morphology of 3 Y-TZP ceramic specimens(3 Y-TZP group),TiO2 thin film-3 Y-TZP ceramic specimens(TiO2 film group),and La-TiO2 thin film-3 Y-TZP ceramic specimens(La-TiO2 film group)wasobserved by scanning electron microscopy,and photocatalysis,antibacterial and cytotoxicity experiments were carried out.For the photocatalytic experiment,four specimens were randomly selected from the La-TiO2 film group and TiO2 film group to observe the degradation rate of methylene blue at different time points under sunlight.In the antimicrobial experiment,five specimens were randomly selected from the La-TiO2 film group,TiO2 film group and 3 Y-TZP group.The antimicrobial activity of each group was tested using the bacteriostatic circle method,and the bacteriostatic distance of each group was compared.In the cytotoxicity experiment,three samples were randomly selected from the LaTiO2 film group,TiO2 film group and 3 Y-TZP group(negative control group),and the extracts were prepared.Here,0.064% phenol solution were served as the positive control group.Morphological observation of L-929 cells in four groups was performed after 3d of culture.Cell viability and relative cell proliferation were measured by MTT assay.Results La-TiO2 films with uniform distribution can be prepared on a 3 Y-TZP surface by the dipping-drawing method.Photocatalytic experiments showed that the degradation rate of methylene blue in the La-TiO2 film group was(41.2±1.5)%
