摘 要:目的探讨血府逐瘀丸对双侧颈总动脉永久结扎所致慢性脑缺血大鼠的保护作用及对脑部NOX表达的影响。方法将50只SD大鼠随机分为空白组、模型组、血府逐瘀丸低剂量组、血府逐瘀丸中剂量组及血府逐瘀丸高剂量组,每组10只。除空白组进行假手术外,其余组均采用双侧颈总动脉永久性结扎法进行慢性脑缺血造模,于造模后第3天,血府逐瘀丸低、中、高剂量组分别给予血府逐瘀丸0. 93 g/kg,1. 87 g/kg,3. 74 g/kg灌胃,1次/d,连续21 d。末次给药24 h后,采用Morris实验检测各组大鼠的学习记忆能力,并对各组大鼠海马体进行尼氏染色及TUNEL染色,采用免疫组化法检测各组大鼠海马体中NOX4表达情况并进行半定量分析。结果训练第2—5天,模型组大鼠逃避潜伏期均明显长于空白组(P均<0. 05),血府逐瘀丸各剂量组均明显短于模型组(P均<0. 05);第6天,模型组大鼠第3象限活动时间和穿越平台次数均明显低于空白组(P均<0. 05),血府逐瘀丸各剂量组均明显高于模型组(P均<0. 05)。血府逐瘀丸各剂量组大鼠海马体中神经细胞核核仁不清晰及细胞形态异常程度均明显轻于模型组,仅有呈散在的少量阳性染色神经元,海马体中NOX4阳性表达均明显弱于模型组,且NOX4相对表达量均明显低于模型组(P均<0. 05)。结论血府逐瘀丸对双侧颈总动脉永久性结扎所致慢性脑缺血大鼠具有较好的神经保护作用,有效抑制海马体中NOX4的表达可能是其主要机制。Objective It is to study the protection effects of Xuefuzhuyu pill on two vesselocclusion induced chronic cerebral ischemia rats and its influence on the expression of NOX4 in brain.Methods 50 SD rats were divided into blank group,model group,low,middle and high dose of Xuefuzhuyu pill groups,each group had 10 rats.Except for the blank group for sham operation,the other groups were treated with permanent bilateral ligation of the common carotid artery for establishing the models of chronic cerebral ischemia.On the third day after modeling,the lower,middle and high dose of Xuefuzhuyu pill groups were respectively given Xuefu Zhuyu pills 0.93 g/kg,1.87 g/kg,3.74 g/kg by lavage,once daily for 21 days.After 24 hours of the last administration,the learning and memory abilities of the rats in each group were detected by Morris test.The hippocampus of each group were stained with Nissl staining and TUNEL.The expression of NOX4 in the hippocampus of each group was detected by immunohistochemical method and a semi-quantitative analysis was performed.Results On the 2nd to 5th day of training,the escape latency of the model group was significantly longer than that of the blank group(P<0.05),and that of every Xuefuzhuyu pill group were significantly shorter than the model group(P<0.05).On the 6th day,the activity time of the third quadrant and the number of crossing platforms in the model group were significantly lower than those in the blank group(P<0.05),and that of Xuefuzhuyu pill groups were significantly higher than those in the model group(P<0.05).The nucleus nucleus and the abnormal morphology of the nucleus in the hippocampus of Xuefuzhuyu pill groups were significantly lighter than those in the model group,only a few positive staining neurons were scattered,and the positive expression of NOX4 in the hippocampus was less than the model group,and the relative expression of NOX4 was significantly lower than that of the model group(P<0.05).Conclusion Xuefuzhuyu pill has a good neuroprotective effect on chronic cerebral
