摘 要:目的通过分析人低分化腺癌、中分化腺癌及癌旁远端正常组织的差异蛋白,寻找与肺腺癌发病相关的蛋白。方法利用双向凝胶电泳的方法分离肺腺癌及癌旁组织的蛋白质,通过图像扫描寻找差异蛋白点,进而应用基质辅助激光电离飞行时间质谱得到肽质量指纹图谱,对比数据库确定差异蛋白。结果人中分化腺癌及癌旁远端正常组织、低分化腺癌及癌旁远端正常组织的平均蛋白点数分别为1810±167、1704±53、2066±144、1549±33。选取差异2倍以上蛋白点进一步分析,发现与肺腺癌相关的蛋白质共14个。结论双向凝胶电泳技术结合基质辅助激光电离飞行时间质谱可鉴定肺腺癌及癌旁远端正常组织的差异蛋白,为早期诊断肺癌,判断预后奠定基础。Objective To analyze the differential proteins of human poorly differentiated adenocarcinoma,moderately differentiated adenocarcinoma and normal tissues adjacent cancer,so as to find the protein associated with lung adenocarcinoma.Methods Different proteins were separated by using two dimensional gel electrophoresis,and then the different proteins were studied and contrasted by using matrix assisted laser ionization time of flight mass spectrometry and image scanning.Results The test were repeated for three times.The number of average spots for human moderately differentiated lung adenocarcinoma and paired normal bronchial mucosa adjacent to tumors and poorly differentiated tissues were 1810±167,1703±53,2066±144,and 1549±33,respectively.14 proteins associated with lung cancer were distinguished.Conclusion Different proteins can be identified by proteomics analysis.The different proteins can find the basis for early diagnosis and estimate the prognosis of lung cancer.
