摘 要:目的研究山茱萸提取物的体外抗氧化活性。方法采用热水提取、酶-Sevage法除蛋白、不同比例乙醇分级沉淀等方法获得山茱萸多糖提取物;经95%乙醇回流提取,乙酸乙酯、正丁醇萃取,得到乙酸乙酯和正丁醇提取物。并利用DPPH·方法,分别测定了各组分的抗氧化活性。结果山茱萸提取物均具有一定的抗氧化活性,且呈显著的量效关系,山茱萸乙酸乙酯、正丁醇提取物清除自由基的能力大于氧化型谷胱甘肽,而小于还原型谷胱甘肽;山茱萸多糖清除自由基的能力均小于氧化型谷胱甘肽,其中90%醇沉多糖较强。结论山茱萸乙酸乙酯、正丁醇提取物以及山茱萸90%醇沉多糖是天然抗氧化剂的良好来源,可以进一步分离纯化。OBJECTIVE Investigate extracorporeal antioxidant activities of the extractives from cornus officinalis. METHODS The polysaccharides were obtained by distilled with hot water,removing proteins on enzyme-Sevag method,further precipitated with different ethanol ratio.The extractives of ethyl acetate and n-butyl alcohol were obtained by distilled with 95% ethanol,extracted with ethyl acetate and n-butyl alcohol.Meanwhile their antioxidation capacity were measured by DPPH·method. RESULTS The extractives from cornus officinalis have obvious antioxidant capacity and show significant dose-effect relationship.Antioxidant activity of extractives extracted by ethyl acetate and n-butyl alcohol are bigger than GSSG and smaller than GSH;the antioxidant activity of polysaccharide is smaller than GSSG.The polysaccharide precipitated by 90% ethanol has stronger antioxidant activity. CONCLUSION The polysaccharide precipitated by 90% ethanol and the extractives extracted by ethyl acetate and n-butyl alcohol are a kind of good natural antioxidants source,they should be worth separated and purified further.
