摘　　要：目的分析基质金属蛋白酶2(MMP2)、组织金属蛋白酶抑制剂2(TIMP2)及人碱性成纤维细胞生长因子(bFGF)在食管鳞癌中的表达及其临床意义。方法选取2017年3月至2018年3月我院收治的食管鳞癌患者120例为研究对象,以其病理确诊的石蜡标本为观察组,取其癌旁正常组织(距离癌组织0.5～1cm)为对照组,应用逆转录-聚合酶链法(RTPCR)测定其MMP2、TIMP2及bFGF mRNA相对表达量,以Western blotting法检测MMP2、TIMP2及bFGF蛋白表达水平,并分析MMP2、TIMP2及bFGF水平与临床病理参数的关系,采用Spearman相关性分析法分析MMP2、TIMP2及bFGF的相关性。结果观察组MMP2及bFGF mRNA相对表达量高于对照组,而其TIMP2 mRNA相对表达量低于对照组(P<0.05);观察组MMP2、bFGF蛋白表达阳性率明显高于对照组,而TIMP2蛋白表达阳性率低于对照组(P<0.05);食管鳞癌患者中,MMP2、TIMP2、bFGF蛋白表达阳性率与肿瘤浸润深度、分化程度、淋巴结转移、临床分期存在一定关系(P<0.05);相关性分析发现,食管鳞癌患者MMP2、bFGF表达水平与TIMP2表达水平呈负相关(P<0.05),而MMP2与bFGF表达水平呈正相关(P<0.05)。结论 MMP2、bFGF在食管鳞癌患者中高表达,而TIMP2低表达,且三者与患者临床病理参数存在一定关系,可能参与食管鳞癌发生发展。Objective To monitor and analyze the expression and clinical significance of matrix metalloproteinase 2(MMP2),tissue inhibitor of metalloproteinase 2(TIMP2)and human basic fibroblast growth factor(bFGF)in esophageal squamous carcinoma.Methods A total of 120 patients with esophageal squamous carcinoma who were admitted to the hospital from March 2017 to March 2018 were selected as the study subjects.Paraffin-embedded specimens confirmed by pathology were used as the observation group,and normal tissues(0.5 to 1 cm away from cancer tissues)were taken as the control group.The mRNA relative expression levels of MMP2,TIMP2 and bFGF were measured by reverse transcription-polymerase chain reaction(RT-PCR),while the protein expression levels of MMP2,TIMP2 and bFGF were detected by western blotting.The relationship between levels of MMP2,TIMP2 and bFGF and clinicopathological parameters was analyzed and Spearman correlation analysis was performed to analyze the correlation of MMP2,TIMP2 and bFGF.Results The mRNA relative expression levels of MMP2 and bFGF in the observation group were higher than those in the control group,while the mRNA relative expression level of TIMP2 was lower than the control group(P<0.05).The positive expression rates of serum MMP2 and bFGF proteins in the observation group were significantly higher than those in the control group,while the positive expression rate of TIMP2 was lower than the control group(P<0.05).In patients with esophageal squamous carcinoma,the positive expression rates of MMP2,TIMP2 and bFGF proteins were related to the depth of tumor invasion,differentiation degree,lymph node metastasis and clinical stage(P<0.05).The correlation analysis found that the expression levels of MMP2 and bFGF were negatively correlated with the expression level of TIMP2(P<0.05),and the expression level of TIMP2 was positively correlated with the expression levels of bFGF in patients with esophageal squamous carcinoma(P<0.05).Conclusion MMP2 and bFGF are highly expressed while TIMP2 is lowly expresse
