摘 要:目的:探讨D-亮氨酸对变异链球菌生物膜形成的影响。方法:分光光度计比浊法检测20 mmol/L D-亮氨酸对变异链球菌浮游细菌生长的影响;体外构建变异链球菌生物膜模型,通过MTT法评价5、10、20 mmol/L D-亮氨酸对变异链球菌生物膜形成的影响,共聚焦显微镜观察不同浓度D-亮氨酸处理后的生物膜形态结构。结果:20 mmol/L D-亮氨酸不影响变异链球菌浮游细菌生长趋势,从3 h左右进入其对数生长期,在12 h到达平台期。MTT法结果表明浓度为5、10、20 mmol/L D-亮氨酸对变异链球菌生物膜均有抑制作用,且随着D-亮氨酸的浓度增加,变异链球菌形成生物膜的量明显降低(P<0.05)。激光共聚焦显微镜观察D-亮氨酸对变异链球菌生物膜作用后,其生物膜结构整体分布稀疏,厚度减少。结论:D-亮氨酸对变异链球菌生物膜形成具有抑制作用。Objective:To investigate the effect of D-leucine on biofilm formation by Streptococcus mutans.Methods:The effect of 20 mmol/L D-leucine on growth curve of Streptococcus mutans was evaluated by the turbidity method.Streptococcus mutans biofilm model was constructed in vitro.Biofilms were formed with 5,10 and 20 mmol/L D-leucine,and the biofilm formation was determined by MTT assay method.Confocal laser scanning microscope(CLSM)was applied to observe the biofilm structure after treated with different concentrations of D-leucine.Results:D-leucine did not affect the growth curves of planktonic Streptococcus mutans within 24 h.The growth curves attained exponential phase after 3 h and reached stationary phase at 12 h.The quantity of biofilm formed by Streptococcus mutans significantly decreased(P<0.05)and the structure of the biofilm showed by CLSM became thinner with the increase of D-leucine concentration.Conclusions:D-leucine can inhibit the biofilm formation of Streptococcus mutans.
