摘　　要：目的探讨C-X-C家族趋化因子受体4(CXCR4)和基质细胞衍生因子1(SDF1)在胃癌中的表达情况及其临床意义。方法选取300例胃癌患者的病理组织标本,同时选取上述胃癌患者的癌旁组织、正常胃组织作为对照。采用免疫组织化学染色法检测CXCR4和SDF1蛋白的表达情况,采用实时逆转录聚合酶链反应(RTPCR)法检测CXCR4 mRNA和SDF1 m RNA的相对表达水平。结果胃癌组织和癌旁组织中的CXCR4蛋白阳性表达率和CXCR4 mRNA相对表达水平均高于正常胃组织,差异均有统计学意义(P﹤0.05)。正常胃组织、癌旁组织和胃癌组织中SDF1蛋白的阳性表达率和SDF1 m RNA相对表达水平比较,差异均无统计学意义(P﹥0.05)。不同临床分期和淋巴结转移胃癌患者胃癌组织中CXCR4蛋白阳性表达率和CXCR4 mRNA相对表达水平比较,差异均有统计学意义(P﹤0.05)。结论 CXCR4蛋白和CXCR4 mRNA相对表达水平在胃癌组织中明显升高,可能与胃癌患者的临床分期和淋巴结转移有关。Objective To investigate the expression of C-X-C motif chemokine receptor 4(CXCR4)and stromal cellderived factor 1(SDF1)in gastric cancer,and to explore the clinical significance.Method A total of 300 patients with gastric cancer were included in the study for retrospective analysis,the gastric cancer tissues,adjacent tissues and normal gastric tissues were collected,the expression of CXCR4 and SDF1 were detected by immunohistochemical staining,and the relative expression of CXCR4 mRNA and SDF1 mRNA were detected by reverse transcription polymerase chain reaction(RT-PCR).Result The positive expression rates of CXCR4 protein and relative expression of CXCR4 mRNA in gastric cancer tissues and adjacent tissues were significantly higher than that in normal gastric tissues(P<0.05).There was no significant difference in the positive expression rate of SDF1 protein and the relative expression level of SDF1 mRNA between normal gastric tissue,adjacent tissue and gastric cancer tissue(P>0.05).While clinical staging and lymph node metastasis were associated with the positive expression of CXCR4 protein and the relative expression of CXCR4 mRNA,with statistically significant differences observed(P<0.05).Conclusion The expression of CXCR4 protein and CXCR4 mRNA in gastric cancer tissues are increased significantly,which may be related to the clinical stage and lymph node metastasis.
