老龄化对单侧肾切除术后大鼠青霉素排泄的影响

摘　　要：目的探究老龄大鼠单侧肾切除术后抗生素类药物排泄的规律和肾功能的变化。方法选用健康雄性Sprague-Dawley大鼠,分为青龄组(3月龄)和老龄组(18月龄),行右肾切除术。术后给两组腹腔注射青霉素,60 min后采血。管碟法检测青霉素血药浓度;试剂盒检测肌酐尿素氮水平评估肾功能;肾脏组织苏木素-伊红(HE)染色观察其病理变化。结果术后老龄大鼠对青霉素的排泄减缓;青、老龄组大鼠术后肾功能生化指标较术前无明显改变;术后10 d时青、老龄组大鼠肾脏质量均代偿性增加;术后老龄组肾小管及间质慢性炎症和纤维化强于青龄组。结论老龄大鼠在术后发生了明显的病理生理特征性改变从而导致青霉素的排泄减缓,但其肾功能无明显改变。Objective To investigate the excretion of frequently used antibiotics and the changes of renal functions of the aged rats after unilateral nephrectomy and provide evidences for clinical medication and further study.Methods Male Sprague-Dawley(SD)rats were allocated into young nephrectomy group(3 months)and age nephrectomy group(18 months).Blood was collected at 60 min after traperitoneal injecting penicillin.Cylinder-Plate Method was used to assess serum penicillin levels.Serum creatinine and urea nitrogen levels were assessed to evaluate renal functions.HE staining of kidney tissues was used to observe histopathologic changes.Results The excretions of serum penicillin of the aged rats were decreased after the surgery.The renal functions of the aged rats before surgery and 10 days after surgery were not significantly different.Kidney weights were increased after the surgery both in young and aged rats.Chronic inflammation and fibrosis in the renal tubules and interstitial of the aged group were more severe than those of young groups.Conclusions Significant pathophysiological changes exhibit in the aged rats with unilateral nephrectomy,which lead to the decrease of penicillin excretion.
