摘 要:目的探讨HIV-1辅助受体基因预测工具对我国流行的CRF01_AE毒株辅助受体利用的可行性。方法从北京市、广西壮族自治区、四川省的HIV-1感染者中分离出CRF01_AE毒株22株,利用基因型方法(11/25规则、WebPSSM和geno2pheno)对病毒辅助受体利用情况进行预测,将预测结果与经GHOST细胞系检测的表型实验结果进行比较。结果经GHOST细胞系检测,22株CRF01_AE毒株中,4株利用CCR5/CXCR4辅助受体,18株利用CCR5辅助受体。11/25规则预测与表型实验的一致率为86.4%;geno2pheno中,G2p-clinical、NGS-sanger和G2p-colon模型预测与表型实验的一致率分别为95.5%、86.4%和68.2%;WebPSSM中,PSSM-sinsi_B、PSSM-x4r5和PSSM-sinsi_C模型预测与表型实验的一致率分别为86.4%、72.7%和45.5%。结论对于我国CRF01_AE毒株的HIV-1感染者,在使用辅助受体拮抗剂之前应进行辅助受体利用的预测,Geno2pheno是较准确的辅助受体基因预测工具。Objective To analyze feasibility of using genotypic prediction for coreceptor usage of HIV-1 CRF01_AE strains circulating in China. Methods Twenty-two CRF01_AE strains were isolated from HIV-1 infected patients in Beijing, Guangxi and Sichuan. The genotypic methods (11/25 rule, WebPSSM and geno2pheno) were used to predict coreceptor usage of the strains. The prediction were compared with the results of phenotypic testing based on Ghost cell line. Results Of the 22 CRF01_AE strains, 4 strains used CCR5/CXCR4 coreceptor and 18 strains used the CCR5 coreceptor, as detected by the GHOST cell line test. The consistency rate between genotypic prediction by 11/25 rule and phenotypic test was 86.4%. For geno2pheno, the consistency rates between genotypic predictions by G2p-clinical, NGS-sanger and G2p-colon models and phenotypic tests were 95.5%, 86.4% and 68.2%, respectively. For WebPSSM, the consistency rates between genotypic predictions by PSSM-sinsi_B, PSSM-x4r5, and PSSM-sinsi_C models and phenotypic tests were 86.4%, 72.7% and 45.5%, respectively. Conclusions For the patients infected with HIV-1 CRF01_AE strains in China, coreceptor usage should be predicted before treatment of coreceptor antagonists. Geno2pheno is a more accurate way for genotypic prediction of HIV coreceptor usage.
