摘 要:为了探究平菇多糖的提取工艺,本试验选用甘露聚糖酶、木瓜蛋白酶、纤维素酶作为提取平菇多糖的复合酶,通过正交设计试验优化复合酶用量,采用响应面优化法考察液料比、酶解温度、酶解时间、pH值四个因素对多糖提取率的影响。结果显示,复合酶的最佳用量分别为甘露聚糖酶400 U/g,木瓜蛋白酶240 U/g,纤维素酶600 U/g,最佳提取工艺条件为液料比21∶1 (mL/g),酶解温度52.0℃,酶解时间3.1 h,pH值5.7验证试验,多糖提取率为5.90%,纯度60.91%;相比之下,热水浸提法多糖提取率为5.72%,纯度59.71%。因此,复合酶法提取可作为一种平菇多糖适宜的提取方法。In order to investigate the extraction process of Pleurotus ostreatus polysaccharides, mannanase, papain, and cellulase were used as a multi-enzyme to extract Pleurotus ostreatus polysaccharides in this study. The orthogonal design test was used to optimize the amount of different enzyme dosages. The response surface optimization method was employed to investigate the effect on the polysaccharide extraction rate by four factors: the ratio of liquid to material, the temperature of enzymolysis, the time of enzymolysis and the pH value. The verification tests were then carried out and comparison was made with hot water extraction. The results showed that the optimum extraction enzyme formula was mannanase 400 U/g, papain 240 U/g and cellulase 600 U/g. The optimum extraction conditions were liquid to material ratio 21 ∶ 1 (mL/g), the enzymolysis temperature at 52℃, the enzymolysis time of 3.1 h and pH value of 5.7. The result of the verification tests showed that the extraction rate of polysaccharides by the multi-enzyme method was 5.90 % with a purity of 60.91 %, while the extraction rate of polysaccharides by the hot water extraction was 5.72 % with a purity of 59.71 %. The study proved that the multi-enzyme method could be used as a suitable extraction method for Pleurotus ostreatus polysaccharides.
