摘  要:目的对片姜黄挥发油成分进行定性定量分析,明确片姜黄挥发油的主要成分及其中10种成分的含量,为进一步研究片姜黄,控制片姜黄质量提供科学依据。方法采用气相色谱-质谱联用、气相色谱法进行定性定量分析,使用Agilent 19091S-433(0.25μm×0.25 mm×30 m)及Elite-WAX ETR(0.25μm×0.25 mm×30 m)色谱柱;程序升温:初始温度70℃,保持2 min,以6℃·min^(-1)升至150℃,以8℃·min^(-1)升至230℃,保持15 min;载气流量2.0 mL·min^(-1);分流进样,进样口温度:250℃,进样量1.0μL。GC-MS用载气为高纯氦气(≥99.999%),质谱条件为EI源,轰击电压70 eV,正离子模式,质量扫描范围50~550 amu,离子源温度230℃;GC用载气为高纯氮气(≥99.999%),检测器为FID,检测器温度为280℃。结果分析12个批次的片姜黄挥发油样品后,鉴定了44种成分,并对其中的10种成分进行了GC定量分析,定量结果准确可靠。结论片姜黄挥发油成分复杂,其中主要组成成分为莪术二酮、樟脑、β-榄香烯、异龙脑等物质。气相色谱定量法简单可靠,可用于片姜黄挥发油的质量控制。Objective To qualitatively and quantitatively analyze volatile oil constituents of Rhizoma Wenyujin Concisum and to determine the main constituents and 10 content of essential oil. To provide a scientific basis to control the quality of Rhizoma Wenyujin Concisum. Methods Qualitative and quantitative analysis was performed by gas chromatography-mass spectrometry and gas chromatography with Agilent 19091 S-433(0.25μm×0.25 mm×30 m) and Elite-WAX ETR(0.25 μm×0.25 mm×30 m) as the columns. Program warming:initial temperature at 70 ℃ for 2 min, rising at 6 ℃·min^-1 to 150 ℃, and then at 8 ℃·min^-1 to 230 ℃ for15 min;carrier gas flow at 2.0 mL·min^-1;split injection, with the inlet temperature at 250 ℃;and injection volume at 1.0 μL. GC-MS carrier gas was high purity helium(≥ 99.999%), mass spectrometry conditions for the EI source, bombardment voltage at 70 eV, positive ion mode, ion source temperature at 230 ℃;GC carrier gas was high purity nitrogen(≥ 99.999%), the detector was FID, and the detector temperature at 280 ℃.Results After analyzing 12 batches of Rhizoma Wenyujin Concisum volatile oil samples, 44 substances were identified and 10 of them were analyzed by GC. The quantitative results were accurate and reliable. Conclusion The volatile oil composition of Rhizoma Wenyujin Concisum is complex, and the main components include curdione, camphor,β-elemene, isoborneol and other substances. The gas chromatography is simple and reliable,and can be used to control the quality of volatile oil and other preparations of Rhizoma Wenyujin Concisum.
