摘 要:目的研究miRNA210在喉癌多药耐药中的调控机制。方法采用荧光素酶报告基因载体实验验证并确定了miRNA210的靶基因,进而构建miRNA210抑制物载体,通过抑制miRNA210的表达,观察其对喉癌多药耐药的调控作用。结果 miRNA210靶基因为NUPR1,抑制miRNA210的表达,可明显增加喉癌细胞对化疗的敏感性,miRNA210可通过调控其靶基因的表达影响喉癌的多药耐药性。结论 miRNA210表达参与喉癌多药耐药的调控机制,以miRNA210为靶点的药物开发及逆转治疗有潜在应用前景。Objective To study the regulation mechanism of miRNA-210 in multidrug resistance of laryngeal cancer.Methods luciferase reporter gene vector test were used to identify the target genes of miRNA-210.Next,in order to observe the regulation effects of miRNA-210 on multidrug resistance(MDR)in laryngeal cancer,the miRNA-210 inhibitor vector was constructed to inhibit the expression of miRNA-210.Results Target gene of the miRNA-210 is NUPR1.It can significantly increase the sensitivity of laryngeal cancer cells to chemotherapy by inhibition of miRNA-210 expression.MiRNA210 can affect the multidrug resistance of laryngeal carcinoma by regulating the expression of its target genes.Conclusion The expression and regulatory of miRNA210 involved in MDR mechanism of in laryngeal carcinoma.Drug development and reversal therapy with targeting miRNA210 have potential application prospects.
