摘  要:目的筛选何首乌有效组分降血脂的配伍方案,探讨胆固醇代谢相关酶的可能机制。方法以大黄素(a)、大黄酸(b)、芦荟大黄素(c)为三个因素,设8个水平,参考均匀设计U_8~3表,设计出A~H八种组方,通过高脂饲料喂养+高脂乳剂(10mL/kg)4周的方法建立大鼠高脂血症模型,分为对照组、模型组及各配方组共10组,在给药4周后,检测血脂水平,结合综合评分和逐步回归分析筛选最优配比方案;用高脂饲料建立SD大鼠高脂血症模型,设对照组、模型组、蒽醌组(60mg/kg)、优选配方组(0.8g/kg,0.04g/kg)、血脂康胶囊组(0.6g/kg)5个组,给药6周后取材,检测血清TC,采用ELISA和RT-PCR法检测HMG-CoA还原酶、CYP7A1的含量及mRNA的表达。结果与模型组比较A、B、C、D、E、G组均能显著降低TC(P<0.05),优选配方组TC的含量较模型组明显降低(P<0.05);HMG-CoA在高脂血症大鼠肝组织中含量明显增多,mRNA表达有增强趋势,优选组方能降低其含量。结论何首乌有效组分优选配方通过抑制HMG-CoA还原酶mRNA的表达水平及其含量;增加CYP7A1mRNA表达水平及其含量,具有抑制胆固醇在体内合成和促进胆固醇转化的双重作用,以达到降血脂的作用。Objective To screen the compatibility of the active components of Polygonum multiflorum to reduce blood lipids,and explore the possible mechanism of cholesterol metabolism related enzymes.Methods First,emodin(a),rhein(b),aloe-emodin(c)as three factors,set 8 levels,refer to the uniform design U8^3 table,design A-H eight kinds of prescriptions.Establishment of rat hyperlipidemia model by high-fat diet + high-fat emulsion(10 mL/kg)for 4 weeks.Divided into control group,model group and each formula group,a total of 10 groups,after 4 weeks of administration,blood lipid levels were measured.Combining comprehensive scores and stepwise regression analysis to screen the optimal ratio scheme;Second,establish a hyperlipidemia model of SD rats with high fat diet.Five groups including control group,model group,anthraquinone group(60 mg/kg),preferred formula group(0.8 g/kg,0.04 g/kg)and Xuezhikang capsule group(0.6 g/kg).After 6 weeks of administration,serum TC was taken,and the contents of HMG-CoA reductase,CYP7A1 and mRNA were detected by ELISA and RT-PCR.Results Compared with the model group,A,B,C,D,E,and G groups could significantly reduce TC(P<0.05).The content of TC in the preferred formula group was significantly lower than that in the model group(P<0.05);HMG-CoA was the content of liver tissue in hyperlipidemia rats increased significantly,and the mRNA expression increased.It is preferred that the group can reduce its content.Conclusion The optimal formulation of Polygonum multiflorum Thunb.inhibits the expression level and content of HMG-CoA reductase mRNA.Increasing the expression level and content of CYP7A1 mRNA can inhibit the synthesis of cholesterol and promote cholesterol conversion in order to achieve hypolipidemiceffect.
