摘 要:在活化培养放线菌A45时,发现原平板上放线菌有被抑制现象,通过进一步分离培养及接种,确定其确有抑制放线菌A45的能力。为了研究该菌的抑菌效果,通过有机试剂乙酸乙酯萃取该菌发酵液,制备滤纸片,发现该菌发酵液对甲型副伤寒杆菌和表皮葡萄球菌抑菌能力较好。通过微量肉汤稀释法测定其最低抑菌浓度,可得该菌产生的抑菌成分对表皮葡萄球菌、伤寒杆菌、甲型副伤寒杆菌、痢疾杆菌、金黄色葡萄球菌的最低抑菌浓度为8mg/mL,对枯草芽孢杆菌和大肠杆菌的抑菌浓度为16mg/mL。最后通过薄层层析最佳流动相的测定探索出两个较好的系统,分别为甲醇:正丁醇:冰乙酸:蒸馏水=9:7:1:2(体积),正丁醇:乙醇:蒸馏水=3:7:7(体积)。When activated culture of Actinomycete A45, it was found that the Actinomycete was inhibited on the original plate. Further isolation, culture and inoculation confirmed that it had the ability to inhibit Actinomycete A45. In order to study the bacteriostatic effect of the bacteria, the fermentation liquid of the bacteria was extracted with organic reagent ethyl acetate, and filter paper was prepared. It was found that the fermentation liquid of the bacteria had better bacteriostatic ability against Salmonella paratyphi A, and Staphylococcus epidermidis. By determining its minimum inhibitory concentration with Microbroth dilution method, it can be obtained that the minimum inhibitory concentration of the bacteriostatic components produced by the bacteria is 8mg/mL for Staphylococcus epidermidis, Typhoid bacillus, Salmonella paratyphi A, Dysentery bacillus and Staphylococcus aureus, and 16mg/mL for Bacillus subtilis and Escherichia coli.Finally, two better systems were found by the determination of the optimal mobile phase by thin layer chromatography: methanol: n-butanol: glacial acetic acid: distilled water =9:7:1:2(volume), and n-butanol: ethanol: distilled water=3:7:7(volume).
