摘 要:背景:目前,临床上对于骨肉瘤(OS)的治疗方法以新辅助化疗联合根治性手术治疗为主,虽然化疗提高了患者的5年生存率,但是化疗药物均具有较高的毒副作用。目的:探讨葡萄籽原花青素(PC)对人骨肉瘤MG63细胞增殖的抑制处理MG63细胞,采用MTT法检测葡萄籽PC对MG63细胞增殖的抑制情况;流式细胞术检测MG63细胞周期;Western印迹法检测葡萄籽PC激酶CDK1和CDK2,细胞周期蛋白依赖性激酶抑制因子p21和p27表达水平的调控。结果:MTT结果显示葡萄籽PC在30~90μg/ml浓度范围内抑制MG63细胞增殖,这种抑制作用具有浓度和时间依赖性;流式细胞术结果显示该细胞被阻滞在G_2/M期和S期(P<0.05);Western印迹结果显示随着葡萄籽PC浓度升高,p21和p27水平升高,cyclinA、cyclin B1、CDK1和CDK2水平降低。结论:葡萄籽PC呈时间和浓度依赖性抑制MG63细胞增殖,其机制可能与将细胞阻滞于G_2/M期和S期有关。Background:At present,the clinical treatment of osteosarcoma(OS)is mainly neoadjuvant chemotherapy combined with radical surgery.Although chemotherapy improves the 5-year survival rate of patients,chemotherapy drugs have high toxicity and side effects.Objective:To investigate the inhibition of grape seed extract proanthocyanidins(PC)on the proliferation of human osteosarcoma cell line MG63 and its mechanism.Methods:MG63 cells were treated with different concentrations of grape seed extract PC.The inhibition of grape seed extract PC on proliferation of MG63 cells was detected by MTF assay.The cell cycle of MG63 was detected by flow cytometry.The expression of cyclin A and cyclin B1,cyclin-dependent kinases CDK1 and CDK2,cyclin-dependent kinase inhibitors p21 and p27 were detected by Western Blot.Results:MTT assay showed that grape seed extract PC inhibit MG63 cell proliferation in the concentration range of 30-90μg/ml.The inhibition was concentration-and timedependent.Flow cytometry showed that the cell was arrested in G2/M and S phases(P<0.05);Western Blot results showed that with the increase of grape seed extract PC concentration,the levels of p21 and p27 increased,and the levels of Cyclin A,cyclin Bl,CDK 1 and CDK2 levels decreased.Conclusions:grape seed extract PC can significantly inhibit the proliferation of MG63 cells in a time-and concentration-dependent manner,which may be related to the arrest of cells in G/M phase and S phase.
