摘 要:目的探讨KAI1基因在不同分化程度胃腺癌细胞株中的表达情况,比较不同胃腺癌细胞株体内、外生长及迁移能力之间的差异。方法选取3种不同分化人胃腺癌细胞BGC823(低分化)、SGC7901(中分化)、MKN28(高分化),采用Western blot及RT-PCR方法检测3种细胞中KAI1蛋白及mRNA表达情况,并应用四甲基偶氮唑盐比色法(MTT)检测3种细胞的生长情况、流式细胞术检测细胞周期的差异、Transwell实验检测细胞迁移的情况。构建3种胃腺癌细胞裸鼠腹腔移植瘤模型,观察其体内生长、迁移情况。结果人胃腺癌BGC823、SGC7901和MKN28细胞中,KAI1蛋白及mRNA的相对表达量呈依次递增趋势(P<0.05)。BGC823、SGC7901和MKN28细胞的24 h相对增殖率呈逐渐下降的趋势(P<0.05),MKN28细胞48 h相对增殖率显著低于BGC823、SGC7901细胞(P<0.05)。BGC823、SGC7901和MKN28细胞S期比例也呈逐渐下降的趋势(P<0.05),且BGC823细胞S期比例最高,DNA合成最多,增殖能力最强。BGC823、SGC7901和MKN28细胞的迁移数目依次呈下降的趋势(P<0.05)。3组裸鼠腹腔移植瘤模型均构建成功,BGC823组、SGC7901组和MKN28组裸鼠腹腔肠系膜移植瘤数目、重量以及腹腔移植瘤转移脏器数均呈依次递减趋势(P<0.05)。结论 KAI1基因的表达可能与胃腺癌细胞的生长、迁移能力有关,KAI1基因表达高的胃腺癌细胞株,其生长、迁移能力弱,反之则生长、迁移能力强。Objective To investigate the expression of KAI1 gene in gastric adenocarcinoma cell lines with different degrees of differentiation,and the variances in growth and migration ability of three gastric adenocarcinoma cell lines in vitro and in vivo . Methods Three different differentiated human gastric adenocarcinoma cells BGC823 (low differentiation),SGC7901 (medium differentiation) and MKN28 (high differentiation) were selected.The protein and mRNA expression of KAI1 in three kinds of cells were detected by Western blot and RT-PCR.The growth of three kinds of cells was detected by MTT assay,the cell cycle was detected by flow cytometry,and the cell migration was detected by Transwell assay.Three nude mice models of gastric adenocarcinoma cells were constructed and their growth and migration were observed. Results The relative expression of KAI1 protein and mRNA in human gastric adenocarcinoma BGC823,SGC7901 and MKN28 cells increased in turn ( P <0.05).The relative proliferation rate of BGC823,SGC7901 and MKN28 cells decreased gradually at 24 h ( P <0.05),and the relative proliferation rate of MKN28 cells at 48h was significantly lower than that of BGC823 and SGC7901 cells ( P <0.05).The S phase ratio of BGC823,SGC7901 and MKN28 cells decreased gradually ( P <0.05),and BGC823 cells had the highest proportion of S phase,the most DNA synthesis,and the strongest proliferation ability.The number of migration of BGC823,SGC7901 and MKN28 cells showed a decreasing trend ( P <0.05).The three groups of nude mice were successfully constructed.The number and weight of peritoneal mesenteric tumors in the BGC823 group,SGC7901 group and MKN28 group and the number of metastatic organs in the abdominal cavity were decreasing ( P <0.05). Conclusion The expression of KAI1 gene may be related to the growth and migration ability of gastric adenocarcinoma cells.The gastric adenocarcinoma cell line with high KAI1 gene expression has weak growth and migration ability,otherwise the growth and migration ability is strong.
