摘  要:目的探讨人脐血间充质干细胞(MSC)移植对大鼠脑缺血再灌注损伤的保护作用及其与SIRT1-P53通路的关系。方法选择40只健康雄性SD大鼠,随机均分为4组:假手术组、模型组、干细胞组、干细胞+EX527组,每组10只。采用逆转录-聚合酶链反应(RT-PCR)法和Western blot法检测各组大鼠不同时间点(1、7、14 d)脑组织匀浆中SIRT1、P53、BAX mRNA含量及蛋白表达变化。于1、3、7、14d对大鼠进行神经功能缺损评分。结果干细胞组与模型组、干细胞+EX527组比较,3、7、14 d神经功能缺损评分明显降低(P <0. 01)。干细胞组SIRT1 mRNA及蛋白表达量明显高于模型组和干细胞+EX527组(P <0. 01),P53、BAX mRNA明显低于模型组和干细胞+EX527组(P <0. 01)。结论人脐血间充质干细胞移植对大鼠脑缺血再灌注损伤的保护机制可能与激活SIRT1-P53通路、下调P53及其下游BAX表达有关。Objective To investigate the protective effect of human umbilical cord blood mesenchymal stem cells(MSC)transplantation on cerebral ischemia-reperfusion injury in rats and its relationship with SIRT1-P53 pathway.Methods 40 healthy male SD rats were randomly divided into 4 groups:sham group,model group,MSC group,and MSC+EX527 group,10 in each group.Reverse transcription-polymerase chain reaction(RT-PCR)and Western blot were used to detect the changes of SIRT1,P53,BAX mRNA expression and protein expression in brain homogenates at different time points(1 d,7 d,14 d),modified neurological severity score(mNSS)were measured 1 d,3 d,7 d and 14 d after reperfusion.Results SIRT1 mRNA and protein expression in the MSC group was significantly higher than that in the model group and the MSC+EX527 group(P<0.01),P53,BAX mRNA expression was significantly lower than model group and MSC+EX527 group(P<0.01).Conclusion The protective mechanism of human umbilical cord blood mesenchymal stem cell transplantation on cerebral ischemia-reperfusion injury in rats may be related to activation of SIRT1-P53 pathway and down-regulation of P53 and its downstream BAX expression.
