摘 要:目的探讨脑原性神经营养因子(brain-derived neuotrophic factor,BDNF)对间歇性主动饮酒大鼠酒精戒断后再饮酒量的影响。方法将42只雄性SD大鼠随机分为空白组(n=6)和模型组(n=36)。空白组无限制自由饮水,模型组间歇性主动饮酒28d后,根据戒断时间分为12h组(n=6)、24h组(n=6)、48h组(n=6)、72h组(n=6)、BDNF组(为戒断72h后VTA区注BDNF,n=6)、人工脑脊液组(为戒断72h后VTA区注人工脑脊液,n=6)。所有大鼠每隔7天同一时间称重一次;检测12h组、24h组、48h组、72h组不同酒精戒断时间后的6h内的再饮酒量;检测BDNF组在戒断72h后VTA区注入BDNF对再饮酒量的影响。结果模型组间歇性主动饮酒28d,饮酒量达到稳定状态,成功建立酒精依赖模型;模型组与空白组比较,大鼠体重无显著性变化(P>0.05); 72h组6h内的再饮酒量明显增高,与12h组、24h组、48h组比较,差异有统计学意义(P<0.05); BDNF组戒断72小时后VTA区注射BDNF再饮酒量显著降低(P<0.05),与72h组和人工脑脊液组具有统计学差异(P<0.05)。结论大鼠间歇性主动饮酒28天成功建立酒精依赖模型,BDNF能显著降低间歇性主动饮酒大鼠酒精戒断后的再饮酒量。Objective To investigate the effect of BDNF (Brain-derived Neuotrophic Factor) on alcohol consumption in intermittent active drinking rats after alcohol withdrawal.Methods Forty-two male SD rats were randomly divided into the blank group (n=6) and the model group (n=36). After 28 days of intermittent active drinking, the model group was divided into 12h group (n=6), 24h group (n=6), 48h group (n=6), 72h group (n=6), BDNF group (n=6) and artificial cerebrospinal fluid group (n=6) according to the withdrawal time.The weight of all rats were measured at the same time every 7 days.The amount of alcohol consumption within 6 hours after different alcohol withdrawal time was detected in groups of 12h, 24h, 48h and 72h.To investigate the effect of BDNF on the amount of re-drinking in VTA after 72 hours of abstinence in BDNF group.Results Model group intermittent active drinking for 28 days, the amount of alcohol consumption reached a stable state, the successful establishment of alcohol dependence model;Compared with the blank group, the body weight of the model group had no significantly change ( P >0.05).The re-drinking amount within 6 hours in the 72h group was significantly higher than that in the 12h group, 24h group and 48h group, and the difference was statistically significant ( P <0.05).After 72 hours of abstinence, the amount of BDNF re-drinking in the VTA area was significantly reduced ( P <0.05), which was statistically different from that in the 72h group and the artificial cerebrospinal fluid group ( P <0.05).Conclusion The alcohol dependence model was successfully established in rats with intermittent active drinking for 28 days, and BDNF could significantly reduce the amount of re-drinking after intermittent active drinking.
