摘 要:目的:探索吉马酮对人非小细胞肺癌NCI-H1770细胞增殖、凋亡、侵袭和迁移的影响。方法:非小细胞肺癌细胞分为4组:对照组(NCI-H1770)、吉马酮(5、10、20μmol/L)组。CCK-8检测细胞增殖。流式细胞术检测细胞凋亡。Transwell检测细胞侵袭。划痕实验检测细胞迁移。蛋白印迹检测细胞增殖核抗原-67(Ki67),Cleaved Caspase-3和血管内皮细胞生长因子(VEGF)的蛋白水平。结果:低浓度(<20μmol/L)的吉马酮不会对非小细胞肺癌细胞产生毒性,高浓度(≥20μmol/L)的吉马酮会急剧减弱非小细胞肺癌细胞活力。与对照组相比,吉马酮(10、20μmol/L)组细胞增殖倍数下降(P <0. 01)。吉马酮(5、10、20μmol/L)组细胞凋亡率高于对照组(P <0. 01)。吉马酮(5、10、20μmol/L)组每个视野下的侵袭细胞数目低于对照组(P <0. 01)。与对照组相比,吉马酮(5、10、20μmol/L)组划痕愈合率降低(P <0. 01)。吉马酮(5、10、20μmol/L)组Ki67和VEGF的相对蛋白水平低于对照组(P <0. 01)。与对照组相比,吉马酮(5、10、20μmol/L)组Cleaved Caspase-3的相对蛋白水平上升(P <0. 01)。结论:吉马酮可降低人非小细胞肺癌NCI-H1770细胞增殖、侵袭及迁移,促进细胞凋亡。Objective: To explore the effects of germacrone on the proliferation,apoptosis,invasion and migration of human non small cell lung cancer NCI-H1770 cells.Methods: The non-small cell lung cancer cells were divided into four groups: control group,Germacrone(5,10,20 μmol/L) groups.The cell proliferation was detected by CCK-8.The apoptosis was measured by flow cytometry.Cell invasion was tested by transwell.Cell migration was detected by wound healing.The protein levels of antigen identified by monoclonal antibody(Ki67),Cleaved Caspase-3 and vascular endothelial growth factor(VEGF) were measured by Western blot.Results: Low concentration(< 20 μmol/L) germacrone had no effect on cell viability of non-small cell lung cancer cells.The cell viability of non-small cell lung cancer cells was significantly decreased by high concentration(≥20 μmol/L) germacrone.Compared with control group,the proliferation folds in Germacrone(10,20 μmol/L) groups were reduced(P < 0.01).Apoptosis in Germacrone(5,10,20 μmol/L) groups was higher than control group(P < 0.01).The number of invasive cells per field in Germacrone(5,10,20 μmol/L) groups was lower than control group(P < 0.01).Compared with control group,the wound closure rate in Germacrone(5,10,20 μmol/L) groups was decreased(P <0.01).The protein levels of Ki67 and VEGF in Germacrone(5,10,20 μmol/L) groups were lower than control group(P < 0.01).Compared with control group,the protein levels of Cleaved Caspase-3 were elevated(P < 0.01).Conclusion: Germacrone inhibites the proliferation,invasion and migration and promotes apoptosis of human non small cell lung cancer NCI-H1770 cells.
