摘 要:目的:调查19个常染色体短串联重复序列(short tandem repeat, STR)基因座在中国苏州地区汉族人群中的遗传多态性,考察其在法医遗传学相关鉴定中的应用价值,并为涉及该地区的群体遗传学研究提供基础数据支持。方法:采用AGCU Expressmarker 20荧光检测试剂盒对1 050名中国苏州地区汉族无关个体进行STR扩增及检测,根据各基因座分型结果计算了法医遗传学相关参数,并分析了苏州汉族与其他地区汉族在遗传距离上的相近程度。结果:经Bonferroni校正后,19个常染色体STR基因座均符合Hardy-Weinberg平衡(P>0.05/19=0.002 6),累积个人识别概率达到0.999 999 999 999 999,二联体累积非父排除概率为0.999 992 521 059 119,三联体累积非父排除概率为0.999 999 997 756 847。多维尺度分析显示苏州汉族同其他地区汉族人群遗传距离基本呈现与地理位置分布相符合的特征。结论 :本研究纳入的19个常染色体STR基因座在中国苏州地区汉族人群中具有较高的遗传多态性,可应用于法医遗传学相关鉴定和群体遗传学研究。Objective : To investigate the genetic polymorphisms of 19 autosomal STR loci in Suzhou Han population and to explore the application values in forensic practice, in order to provide fundamental data for studies concerning population genetics of this region. Methods : A total of 1 050 unrelated Han individual samples in Suzhou were collected and amplified using the AGCU Expressmarker 20 system kit. The forensic parameters were calculated based on the raw genotyping data obtained and the population comparation was conducted based on the genetic distances between different population. Results : No significant deviation from Hardy-Weinberg e quilibrium w as o bserved f or t he g enotype f requency d istribution o f a ll s tudied l oci a fter Bonferroni's correction(P>0.05/19=0.002 6). The cumulative power of discrimination, probability of exclusion for diad and triplet were 0.999 999 999 999 999, 0.999 992 521 059 119 and 0.999 999 997 756 847, respectively. Besides, the multidimensional scaling analysis revealed that the genetic distances between Suzhou Han and other Han population were basically in accordance with their geographic distributions. Conclusion : The 19 autosomal STR loci enrolled in our study exhibited relatively high genetic polymorphisms in Suzhou population, which could imply a strong validity for forensic identification, paternity testing and population genetic research.
