摘　　要：目的:探讨H37Ra菌株感染后小鼠脾脏IL-35 mRNA的表达情况,分析其在结核分枝杆菌(M.tb)感染中的作用与意义。方法:将清洁级C57B/L6小鼠分为对照组、感染4周组和感染8周组,每组10只。采用H37Ra菌株感染后,观察小鼠一般状况与肺组织病理,并检测肺部感染细菌数及脾组织IL-35的mRNA表达水平。结果:H37Ra菌株感染小鼠在不同时段其脾组织IL-35(EBI3亚基和P35亚基)的mRNA表达均高于对照组(P<0.01),感染8周组的脾组织IL-35的mRNA表达水平更高(P<0.01)。结论:结核分枝杆菌H37Ra菌株感染小鼠后脾组织IL-35 mRNA表达水平升高,IL-35表达可能与结核分枝杆菌感染密切相关。Objective: To detect IL-35 mRNA level in the spleen of mice infected with strain H37Ra for investigation of the role of IL-35 mRNA expression in Mycobacterium tuberculosis infection. Methods :C57B/L6 mice were randomly divided into control group,and model group respectively infected with H37Ra via tail vein injection for 4 weeks and 8 weeks( n= 10 for each group).Then the lung and spleen tissues were taken to undergo examination for the histopathological changes in the lung and IL-35 mRNA in the spleen. Results :Expression of P35 mRNA and EBI3 mRNA in the spleen tissue was higher in mice infected with strain H37Ra than those in the control group( P< 0.01),and IL-35 mRNA level was higher by the 8th week after infection( P< 0.01 ). Conclusion :Up-regulated IL-35 mRNA level was seen in the spleen tissue of mice following strain H37Ra infection,suggesting that IL-35 may be involved in Mycobacterium tuberculosis infection.
