摘 要:目的:研究共培养状态下施万细胞(SCs)对唾液腺腺样囊性癌细胞(SACCs)侵袭、迁移能力的影响及可能机制。方法:用ELISA等检测NT-3、TrkC在共培养状态下的表达情况;通过划痕、侵袭、迁移实验和3D共培养实验,评估各组细胞的侵袭与迁移能力。结果:共培养条件下,NT-3,TrkC表达较单独培养明显升高;SACC-83细胞和SCs的侵袭、迁移能力均显著增强;SCs可早期定向趋化迁移至肿瘤。结论:NT-3/TrkC信号通路可能通过介导SCs向肿瘤细胞的迁移来促进SACC嗜神经侵袭的发生。Objective:To investigate potential mechanism of the perineural invasion and motor ability of the Schwann cells(SCs)on the salivary adenoid cystic carcinoma cells(SACCs).Methods:SCs and SACCs were respectively cultured and co-cultured.The expression of NT-3 and TrkC was measured by ELISA.Wound healing test,invasion and motor test and three-dimensional(3 D)co-culture assays were performed to evaluate the motor ability under the interaction of SACC-83 cells and SCs.Results:The expression of NT-3 and TrkC were markedly elevated and the migration,and invasion ability increased significantly when cells were co-cultured.Moreover,SCs firstly migrated to SACC-83 cells before the tumor attacked the nerve.Conclusion:NT-3/TrkC axis may possibly mediate the migration of SCs towards SACC cells to promote the occurrence of PNI.
