摘 要:目的观察动脉粥样硬化(AS)时大鼠心脏微小血管变化及桂枝茯苓胶囊对其影响作用。方法将40只SD大鼠随机分为空白组11只和造模组29只。8周末两组各取2只处死,病理证实造模成功后随机将造模组大鼠分为观察组、对照组、模型组各9只。分别给予桂枝茯苓胶囊、辛伐他汀和卡托普利、生理盐水灌胃,14周末检测心肌微小动脉密度(MMAD)和心肌毛细血管密度(MCD),基质金属蛋白酶-2(MMP-2)、基质金属蛋白酶2特异性组织抑制因子(TIMP-2)、核转录因子-κB(NF-κB)mRNA及蛋白表达。结果模型组MMAD高于空白组、对照组及观察组,差异有统计学意义(P<0.05),MCD低于空白组、对照组及观察组,差异有统计学意义(P<0.05),空白组、对照组及观察组MMAD差异无统计学意义(P>0.05),观察组MCD高于对照组,差异有统计学意义(P<0.05),观察组与空白组、空白组与对照组间MCD差异无统计学意义(P>0.05)。模型组MMP-2 mRNA及蛋白表达水平高于空白组、对照组及观察组,差异有统计学意义(P<0.05),对照组、观察组及空白组间比较,差异无统计学意义(P>0.05)。观察组TIMP-2 mRNA及蛋白表达水平高于空白组、对照组及模型组,差异有统计学意义(P<0.05),对照组和模型组TIMP-2 mRNA及蛋白表达水平均高于空白组,差异有统计学意义(P<0.05),对照组与模型组间差异无统计学意义(P>0.05)。模型组NF-κB mRNA及蛋白表达高于观察组,差异有统计学意义(P<0.05),观察组高于空白组及对照组,差异有统计学意义(P<0.05),空白组与对照组间差异无统计学意义(P>0.05)。MMP-2 mRNA及蛋白表达与MMAD呈正相关(r=0.65、0.41,P<0.05),与MCD呈负相关(r=-0.59、-0.46,P<0.05)。NF-κB mRNA及蛋白表达与MMAD呈正相关(r=0.69、0.71,P<0.05),与MCD呈负相关(r=-0.49、-0.47,P<0.05)。结论 AS大鼠心脏微小血管发生重塑,可能的机制是NF-κB及MMP-2过表达;桂枝茯苓胶囊有抑制NF-κB及MMP-2过表达作用。Objective To observe the changes of heart microvessels in rats with atherosclerosis(AS)and the effect of Guizhi Fuling Capsule(GFC)on it.Methods Forty SD rats were randomly divided into blank group(11 rats)and model group(29 rats).Two rats in each group were executed at the end of the 8th week.After pathological confirmation,the rats in the model group were randomly divided into observation group,control group and model group with 9 rats in each group.GFC,simvastatin combined with captopril and saline were given intragastrically.Immunohistochemistry technology was used for observing the myocardial microarteries density(MMAD),the myocardial capillaries density(MCD)and the protein expressions of matrix metalloproteinase-2(MMP-2),tissue inhibitor of metalloproteinas-2(TIMP-2),nuclear transcription factor-kappa B(NF-κB).RT-PCR technology was used for detecting the mRNA expressions of MMP-2,TIMP-2,NF-κB at the end of 14 weeks.Results The MMAD of model group was higher than that of blank group,control group and observation group,the difference was statistically significant(P<0.05),MCD was lower than that of blank group,control group and observation group,the difference was statistically significant(P<0.05).There was no significant difference in MMAD between blank group,control group and observation group(P>0.05).The MCD of observation group was higher than that of control group,the difference was statistically significant(P<0.05).There was no significant difference in MCD between the blank group and the blank group and the control group(P>0.05).The expression of MMP-2 in model group was higher than that in blank group,control group and observation group(P<0.05).There was no significant difference among control group,observation group and blank group(P>0.05).The expression of TIMP-2 in the observation group was higher than that in the blank group,the control group and the model group(P<0.05).The expression of TIMP-2 in the control group and the model group was higher than that in the blank group(P<0.05).There was no s
