南方红豆杉水提物对NCI-N87胃癌裸鼠移植瘤凋亡作用及机制研究

摘　　要：目的探讨南方红豆杉水提物对HER2阳性人胃癌NCI-N87细胞裸鼠移植瘤的诱导凋亡作用,并初步探索其作用机制。方法建立荷瘤裸鼠模型,将60只BALB/c裸鼠接种人HER2阳性胃癌NCI-N87细胞株并随机分组给药及取瘤,TUNEL法检测移植瘤细胞凋亡,实时荧光定量PCR法检测PI3K、AKT1、mTOR mRNA表达水平。结果①中药低、中、高剂量组均可见到凋亡细胞,凋亡率同对照组比较,差异有统计学意义(P<0.05);中剂量联合组差异性最为显著(P<0.01);②红豆杉各剂量组PI3K mRNA表达均降低(P<0.05),红豆杉联合赫赛汀后明显下调AKT1 m RNA表达,红豆杉中、低剂量组m TOR mRNA表达显著降低(P<0.05)。结论①南方红豆杉水提物可以诱导胃癌细胞的凋亡,联合西药曲妥珠单抗后并未增强其诱导作用;②南方红豆杉水提物可部分下调PI3K、AKT1、m TOR mRNA表达水平。Objective To investigate the apoptosis effect of aqueous extract of Taxus Chinensis Var. Mairei on the transplanted tumor of nude mide with HER2-positive human gastric cancer NCI-N87 cells, and to preliminarily explore its mechanism. Methods A model of tumor-bearing nude mice was established. 60 BALB/c nude mice were inoculated into human HER2-positive gastric cancer NCI-N87 cell line and randomLy grouped for administration and tumor-removing. TUNEL assay was used to detect the apoptosis of transplanted tumor cells. The expression levels of PI3K, AKT1 and mTOR mRNA were detected by real-time fluorescent quantitative PCR. Results ①Apoptotic cells were observed in the low, medium and high dose groups of Chinese medicine, and the apoptosis rate was compared with the control group, and the difference was statistically significant(P<0.05);the difference in the combined middle dose group was the most significant(P<0.01);②The expression of PI3K mRNA in each dose group of Taxus was decreased(P<0.05). After Taxus combined with Herceptin, the expression of AKT1 mRNA was down-regulated significantly. The expression of mTOR mRNA in the middle and low dose groups of Taxus was significantly decreased (P<0.05). Conclusion ①The aqueous extract of Taxus Chinensis Var. Mairei can induce apoptosis of gastric cancer cells. Its combination with Western medicine of trastuzumab does not enhance the induction;②The aqueous extract of Taxus chinensis var. mairei can partially down-regulate the expression levels of PI3K, AKT1 and mTOR mRNA.
