摘 要:目的:研究hyper-IL-15(HIL-15,包含白介素-15及其受体)基因修饰后的NK-92细胞(HIL-15NK-92细胞)是否可以促进NK-92细胞的增殖并增强其对小鼠体内黑色素瘤的杀伤作用。方法:用CCK-8法检测NK-92细胞和HIL-15NK-92细胞增殖的差异;设立空白对照组、PBS组、NK-92治疗组和HIL-15NK-92治疗组4组体内实验,每组4只C57BL/6雄鼠,皮下接种B16-F10黑色素瘤细胞,观察不同组B16-F10黑色素瘤小鼠肿瘤的生长状况及生存时间。结果:HIL-15基因修饰后的NK-92细胞比NK-92细胞增殖活性显著增强;HIL-15基因修饰后的NK-92细胞治疗组小鼠比NK-92细胞治疗组小鼠的生存期显著延长。结论:HIL-15基因修饰可以促进NK-92细胞的增殖并延长黑色素瘤小鼠的生存期。Objective :To investigate whether NK-92 cells modified with hyper-IL-15(HIL-15) gene which was composed of IL-15 and the domain of the IL-15 receptor chain could promote the proliferation of NK-92 cells and enhance their killing effect on melanoma in mice. Methods :CCK-8 assay was used to detect the proliferation differences between NK-92 cells and HIL-15 NK-92 cells. Blank control group, PBS group, NK-92 treatment group and HIL-15 NK-92 treatment group were established. Four C57 BL/6 male mice in each group were subcutaneously inoculated with B16-F10 melanoma cells. The growth of melanoma and the survival time of mice were observed and recorded.Results :Compared with NK-92 cells, the proliferation activity of NK-92 cells after HIL-15 gene modification was enhanced. The survival time of mice in HIL-15 NK-92 treatment group was longer than that in NK-92 treatment group.Conclusion :HIL-15 gene modification can promote the proliferation of NK-92 cells and prolong the survival time of melanoma bearing mice.
