摘 要:目的观察绿茶多酚对缺血-再灌注氧化损伤肾脏的保护作用。方法 SPF级雄性成年SD大鼠30只,随机分为假手术组(Sham组)、缺血-再灌注组(I/R组)、绿茶多酚+缺血-再灌注组(I/R+G组),每组10只。Sham组给予0.9%氯化钠溶液; I/R组和I/R+G组分别给予0.9%氯化钠溶液60 mg·kg^(-1)·d^(-1)和绿茶多酚60 mg·kg^(-1)·d^(-1)灌胃,4周后行缺血-再灌注处理50 min。所有大鼠手术结束后1 h取肾脏标本进行丙二醛(MDA)、超氧化物歧化酶(SOD)、肾损伤分子-1(KIM-1)和细胞凋亡检测。结果 Sham组SOD和MDA分别为(510.67±16.91) U·mg^(-1),(0.02±0.03)nmol·mg^(-1); I/R组SOD和MDA分别为(174.02±9.62) U·mg^(-1),(4.10±0.09) nmol·mg^(-1); I/R+G组SOD和MDA分别为(439.17±8.27) U·mg^(-1),(1.78±0.08) nmol·mg^(-1)。Sham组和I/R+G组SOD明显较I/R组高,而MDA明显较I/R组低; Sham组、I/R组和I/R+G组KIM-1表达评分分别为0.02±0.02,4.52±0.11,1.98±0.09。I/R组KIM-1表达明显较Sham组和I/R+G组高(P<0.05); Sham组、I/R组和I/R+G组凋亡指数分别为0.08±0.08,53.39±2.95,14.86±1.19,Sham组和I/R+G组凋亡指数明显较I/R组低(P<0.05)。结论绿茶多酚对缺血-再灌注氧化损伤肾脏具有较好的保护作用。Objective To observe the protective effect of green tea polyphenols on ischemia/reperfusion injury of rat kidney. Methods A total 30 male Sprague Dawley rats were randomly divided into sham group ( n =10), I/R group ( n =10) and I/R+G group ( n =10), respectively. Both sham group and I/R group were gavaged with 0.9% sodium chloride solution 60 mg·kg ^-1 ·d^-1 per day for 4 weeks. I/R+G group were gavaged with green tea polyphenols 60 mg·kg ^-1 ·d ^-1 for 4 weeks. Then, the ischemia/reperfusion injury model was established. The left kidneys of I/R group and I/R+G group were underwent ischemia/reperfusion for 50 min. Sixty min after operation, left kidneys were harvested for detection of MDA, SOD, KIM-1 and apoptosis index. Results The SOD of Sham group [(510.67±16.91) U·mg ^-1 ] and I/R+G group [(439.17±8.27) U·mg^-1 ]were higher than that of I/R group[(174.02±9.62) U·mg ^-1 , P <0.05]. The MDA of Sham group [(0.02±0.03) nmol·mg ^-1 ] and I/R+G group [(1.78±0.08) nmol·mg ^-1 ] were significantly lower than that of I/R group [(4.10±0.09) nmol·mg ^-1 , P <0.05]. The score of KIM-1 expression in I/R group (4.52±0.11) was higher than that of Sham group (0.02± 0.02 ) and I/R+G group (1.98±0.09)( P <0.05).The apoptosis index in Sham group (0.08±0.08) and I/R+G group (14.86± 1.19 ) were much lower than that of I/R group (53.39±2.95, P <0.05).Conclusion Green tea polyphenols has exerted protective effect on ischemia/reperfusion injury of rat kidney.
