摘 要:目的:通过生物信息学方法,利用基因表达综合数据库(GEO)分析两种种植体表面(纳米/非纳米级)早期附着细胞的基因表达谱。方法:从GEO数据库下载芯片数据(GSE42288),运用R软件对42枚纳米级(纳米组)和42枚非纳米级表面处理(非纳米组)的迷你种植体周附着细胞的基因谱进行差异表达基因筛选,使用DAVID软件分别对差异基因进行GO及KEGG通路富集分析,同时使用STRING和Cytoscape软件进行蛋白质网络互作分析。结果:筛选出110个差异表达基因(上调7个,下调103个)。GO富集分析结果提示,差异基因在免疫反应、炎症反应、趋化因子调节、细胞凋亡通路等生物学过程及白细胞介素(IL)-8受体活化等分子功能中发挥重要作用;KEGG通路分析结果显示,差异表达基因主要在自然杀伤细胞介导的细胞毒作用、破骨细胞分化、抗原加工提呈、趋化因子、细胞因子及其受体相互作用信号通路上发挥作用;GO、KEGG及蛋白互作网络分析筛选出纳米级种植体组主要的下调差异基因为VEGFA、CCL20、IL-8、CXCR1和CXCR2。结论:纳米级种植体更利于早期骨愈合,这可能与VEGFA、CCL20、IL-8、CXCR1和CXCR2基因的下调有关。Objective: To explore the expression profile of early attachment cells on two different implant surfaces (nanometer/non-nanometer level) by Gene Expression Omnibus (GEO) database and bioinformatics. Methods: An array data of GSE42288 was downloaded from GEO database.The differentially expressed genes in two groups were analyzed with R software.Moreover,Gene Ontology annotation and KEGG pathway enrichment analyses for DEGs were performed by DAVID.A protein-protein interaction network was established using STRING and Cytoscape software. Results: A total of 110 DEGs (7 upregulated and 113 downregulated) were identified in the nanoscale features surface samples compared with microrough surface.GO enrichment analysis indicated that these DEGs played an important role in the biological processes such as immune response,inflammatory response,chemokine-mediated signaling pathway,apoptotic process and the molecular functions such as interleukin-8(IL-8) receptor activity.KEGG pathway showed that DEGs mainly played roles in natural killer cell mediated cytotoxicity,osteoclast differentiation,antigen processing and presentation,chemokine signaling pathway and cytokine-cytokine receptor interaction.The major down-regulated differentially expressed genes VEGFA,CCL20,IL-8,CXCR1 and CXCR2 were screened by GO,KEGG analysis and protein interaction network. Conclusion: Nanoscale features surface is more conducive to primary bone healing,which provides a basis for further experiments to verify the mechanism of action of these genes and pathways in osteogenesis between nanoscale features surface implant and bone osseointegration.
