大鼠肝癌细胞Mael、Ddx4 mRNA及蛋白的表达及意义

摘　　要：目的检测大鼠肝癌细胞中蛋白质漩涡同系物基因(Mael)和生殖细胞特异性蛋白4(Ddx4)的表达变化,探讨Mael和Ddx4基因在肝癌发病中的意义。方法将大鼠肝癌细胞CBRH-7919作为肝癌细胞组,正常肝细胞BRL作为正常肝细胞组,采用qRT-PCR法检测Mael、Ddx4基因mRNA表达,采用Western blotting法检测Mael、Ddx4蛋白表达。结果肝癌细胞组Mael、Ddx4 mRNA及蛋白表达均高于正常肝细胞组(P均<0. 01)。结论大鼠肝癌细胞中Mael、Ddx4基因和蛋白表达均明显增高。Objective To investigate the mRNA and protein expression of protein maelstrom homolog (Mael) and DEAD box polypeptide 4 (Ddx4) in the rat hepatoma carcinoma cells (CBRH-7919), and to explore the significance of Mael and Ddx4 genes in the pathogenesis of liver cancer. Methods The mRNA and protein expression levels of Mael and Ddx4 genes were detected in the rat hepatoma carcinoma cells CBRH-7919 ( hepatoma carcinoma cell group) and normal hepatocyte cells BRL (normal hepatocyte cell group) by qRT-PCR and Western blotting. Results Compared with the normal hepatocyte cell group, the mRNA and protein expression of Mael and Ddx4 in the hepatoma carcinoma cell group was higher (both P <0.05). Conclusion The mRNA and protein expression of Mael and Ddx4 in hepatoma carcinoma cells increases significantly.
