摘 要:目的探讨HDAC9蛋白在结肠癌中的表达,并分析其表达状态与临床病理资料之间的关系。方法通过免疫组织化学分析的方法检测结肠癌组织芯片(结肠癌组织99例和癌旁组织80例)中HDAC9蛋白的表达,利用蛋白印迹方法检测15例结肠癌组织和配对癌旁组织中HDAC9蛋白的表达,并通过SPSS 17.0软件统计分析HDAC9在结肠癌中的临床意义。结果免疫组织化学分析表明,HDAC9阳性产物表达于细胞核,结肠癌组织中HDAC9表达水平明显高于癌旁组织(P=0.001)。HDAC9的表达与结肠癌的淋巴结转移和TNM分期相关。生存分析表明HDAC9高表达可缩短结肠癌患者的生存时间(P=0.003)。Cox回归分析证实HDAC9是一个独立的预后标志物(P=0.022)。蛋白印迹结果发现结肠癌组织中HDAC9的表达明显高于癌旁组织(P=0.04)。结论 HDAC9在结肠癌中可能是一个不良的生存预后因子。Objective To investigate the HDAC9 aberrant expression in colon cancer and analyze the relationship between HDAC9 and clinicopathological patameters in human colon cancer. Methods Immunohistonchemistry (IHC) was used to detect the HDAC9 protein levels in tissue microarrays including 99 primary colon cancer tissues and 80 para-carcinoma tissues. Western blot was used to detect the expressions of HDAC9 in 15 cases of colon cancer tissues and 15 para-carcinoma tissues. The relationship of HDAC9 expression with clinicopathologic parameters was evaluated by SPSS 17.0. Results Immuno-histochemical analysis indicated that HDAC9 expression product was located in nucleus, with higher expression levels in colon cancer tissues than in para-cancerous tissues(<0.001). The expression of HDAC9 was associated with regional lymph node metastasis and TNM stage significantly. Survival analysis indicated that high HDAC9 expression shortened the patients’survival time(P= 0.003). A cox regression analysis confirmed HDAC9 to be an independent prognostic marker(P= 0.022). Western blot analysis indicated that HDAC9 expression was significantly higher in colon cancer tissues than in para-carcinoma tissues(P=0.04). Conclusion HDAC9 is a bad survival factor in colon cancer.
