摘  要:目的:探讨睡眠剥夺复合免疫法建立原发免疫性血小板减少症气不摄血证小鼠模型的可行性。方法:Balb/c小鼠分为正常组、模型组,模型组在第1周仅睡眠剥夺,第2、3周睡眠剥夺复合免疫法(腹腔注射抗血小板血清),实验周期为3周。实验结束后,采用血球计数仪检测血小板数量、称量小鼠脾脏重量并计算脾脏指数、流式仪检测T淋巴细胞亚群(CD3^+、CD4^+、CD8^+),此外,观察并检测小鼠一般体征,采用大小鼠抓力测定仪检测抓力,MouseOx Plus小动物无创监护仪检测脉搏幅度,数码照相机采集舌面特征。结果:疾病指标:模型组与正常组相比血小板计数明显降低,注射部位、耳部、足部等有点状或块状出血,脾脏指数明显升高,T淋巴细胞亚群CD3^+、CD4^+、CD8^+均显著降低,且CD4^+/CD8^+明显降低;证候指标:模型组与正常组相比抓力显著降低,脉搏幅度显著降低,舌象R/G值明显降低。结论:睡眠剥夺复合免疫法可以成功建立与临床接近的原发免疫性血小板减少症气不摄血证小鼠模型。Objective: To explore the feasibility of establishing the mice model of primary immune thrombocytopenia with syndrome of Qi failing to control blood by sleep deprivation combined immunoassay. Methods: Balb/c mice were divided into the normal group and the model group.On the model group,sleep deprivation was taken only in the first week and the sleep deprivation combined immunization method( the antiplatelet serum was intraperitoneally injected) were taken in the second and third weeks. The total experimental period was 3 weeks. At the end of the experiment,the periphery platelet amount was measured by a hemocytometer,the spleen was weighed and spleen index was calculated,the T lymphocyte subsets( CD3^+,CD4^+,CD8^+) were detected by flow cytometry. In addition,the general signs of the mice were observed,the grasping force was measured by a Rat/mouse grip tester,the pulse amplitude was detected by a MouseOx Plus small animal non-invasive monitor,and the tongue images were collected by a digital camera. Results: Disease indicators: Compared with the normal group,the periphery platelet amount in the model group was significantly lower,spot or block bleeding was found at the injection site,ears,feet,etc. The spleen index was significantly increased,T lymphocyte subset( CD3^+,CD4^+,CD8^+) were significantly decreased,and CD4^+/CD8^+ was significantly decreased. Syndrome indicators: Compared with the normal group,the grasping force in the model group was significantly reduced,the pulse amplitude was significantly reduced,and R/G of the tongue image was significantly reduced. Conclusion:The method of sleep deprivation combined immunization can successfully establish the mice model of primary immune thrombocytopenia with syndrome of Qi failing to control blood.
