摘  要:本实验以红甜菜叶片为材料,采用Real-time PCR技术,并结合2-△△Ct法处理数据,对不同生长时期甜菜叶片中的红色素合成相关酶基因的相对表达情况进行了分析。结果表明:酪氨酸酶基因在五对真叶期以前相对表达量较高,说明其可能在早期甜菜叶片中红色素的合成起着一定作用,其前期的高表达有助于红色素在叶片中的积累;葡糖基转移酶基因在甜菜生长的各个时期都发挥着作用,而且各个生长时期其相对表达量差异较小,这说明该基因在整个甜菜生长期都在稳定调控红色素合成;4,5-DOPA-双加氧酶基因在叶片生长完成后,其基本不表达。研究明确了甜菜红色素合成相关基因在不同时期相对表达量情况,实验结果为研究红色素在甜菜叶中不同时期合成代谢提供理论基础和依据。In this experiment,the red beet leaves were used as materials,and the expression of betacyanin synthesis related genes was analyzed by using Real-time PCR and 2^-△△Ct method.The results showed that the expression of tyrosinase gene was higher before the five pairs of true leaf stage.It indicated that it might play a role in the synthesis of betacyanin in beet leaves at the early stages of growth,and its high expression at the early stage helps the store of betacyanin in the leaves.The glucosyltransferase gene plays a role in the growth of sugar beet,and the relative expression difference is small in each growth period,which indicates that the gene stably regulates betacyanin synthesis throughout the sugar beet growth period.When all the leaves have grown up,the 4,5-DOPA-dualoxygenase gene is barely expressed.The study clarified the expression of the genes related to betacyanin synthesis in different periods.The experimental results provide a theoretical basis for studying the anabolism of betacyanin at different stages of sugar beet leaves.
