摘 要:目的:探讨川芎嗪对神经-肌肉接头处兴奋传递的影响。方法:取牛蛙50只,随机分为5组(实验组1~4,对照组, n=10),均常规制作坐骨神经-腓肠肌标本。应用BL-420F生物信号采集处理系统同步记录各标本的神经干动作电位、肌细胞动作电位及肌张力,观察不同浓度中药制剂川芎嗪(1mg/ml、2mg/ml、3mg/ml和4mg/ml)对神经肌肉的电活动和机械收缩的影响。结果:实验各组用药前后自身对照及与对照组组间对照,均能观察到,中药川芎嗪能够降低蛙腓肠肌肌张力,具有显著统计学差异(P <0. 01),且呈剂量依赖性。川芎嗪对神经干动作电位的影响不具有统计学意义(P> 0. 05)。而对肌细胞动作电位幅度的影响,亦呈剂量依赖性降低(P <0. 05)。结论:川芎嗪不影响神经干的兴奋传导过程,而可能通过抑制神经-肌肉接头处兴奋传递过程及骨骼肌兴奋-收缩耦联过程而影响腓肠肌机械收缩活动。Objective:To explore the effect of Ligustrazine on excitatory transmission in nerve-muscle junction. Methods:Fifty bullfrogs were randomly divided into 5 groups (experimental groups 1-4,control group),10 in each group. Sciatic Nerve-gastrocnemius muscle specimens were made routinely.BL-420F biological signal acquisition and analysis system was used to record nerve stem action potential,muscle cell action potential and muscle tension synchronously. The effects of tetramethylpyrazine of different concentration(1mg/ml,2mg/ml,3mg/ml and 4mg/ml) on electrical activity and mechanical contraction of nerve and muscle were observed.At the same time,the changes of gastrocnemius contractile tension induced by direct stimulation of gastrocnemius muscle with electrodes were observed under the background of application of 4mg/ml tetramethylpyrazine.Results:Comparisons were conducted among the experimental groups before and after drug-use and between the experimental groups and control group.Results showed that tetramethylpyrazine could decrease the muscular tension of frog gastrocnemius in a dose-dependent manner,which was of significant statistical difference(P<0.01).The effect of Ligustrazine on action potential of nerve trunk was not statistically significant(P>0.05).And its effect on the action potential amplitude of muscle cells were less dose-dependent comparatively(P<0.05).With tetramethylpyrazine injection(4mg/ml)and same time interval,the muscle tension induced by direct stimulation of gastrocnemius muscle decreased gradually(P<0.05).Conclusion:Ligustrazine does not affect the excitation conduction of nerve trunk,but may affect the contraction efficiency of gastrocnemius muscle by inhibiting the excitation conduction process at nerve-muscle junction and the excitation-contraction coupling process of skeletal muscle.
