摘  要:目的探讨氯化钆对肺腺癌细胞株(A549)迁移侵袭能力的影响,以期能够为肿瘤治疗寻找新的线索.方法用氯化钆联合转化生长因子β(transforming growth factor-β,TGF-β)处理的A549细胞,Transwell小室实验检测A549迁移侵袭能力;实时定量PCR检测相关蛋白的mRNA的转录水平.结果 Transwell小室迁移实验结果显示GdCl3能够降低A549细胞的迁移能力(P<0.05);Transwell小室侵袭实验显示氯化钆处理后,A549细胞侵袭能力减弱(P<0.05);实时定量PCR结果显示N-钙粘蛋白,波形蛋白mRNA减少;E-钙粘蛋白mRNA增加(P值均<0.05).结论氯化钆可起到抑制肿瘤细胞迁移侵袭的作用.通过对氯化钆药理作用的深入研究,可为肿瘤提供新的治疗途径.Objective To explore the effect of gadolinium chloride( Gdd3)on the inhibition of the lung adenocarcinoma cell line(A549)migration and invasion. Methods In this experiment,A549 cells were treated with transforming growth(TGF-β)with or without gadolinium chloride. Transwell chamber experiments were used to detect the migration and invasion;Real-time qPCR was used to detect the mRNA level of E-cadher-in,N-cadherin,and vimentin. Results The Transwell chamber experiments showed that gadolinium chloride could inhibit the migration ability of A549 cells(P < 0. 05). Transwell chamber invasion assay showed that the invasion ability of A549 cells decreased after the intervention of gadolinium chloride( P < 0. 05). Real-time qPCR showed that the expression of N-cadherin and vimentin decreased at the mRNA level,while the expression of E-cadherin increased compared with the TGF-β treated group after the intervention of gadolinium chloride(all P < 0. 05). Conclusions Gadolinium chloride could decrease the migration and invasion of A549 cells,which could provide a new way of treatment for lung adenocarcinoma.
