摘　　要：目的:探讨Zebularine对乳腺癌细胞株MDA-MB-231细胞增殖和SARI表达的影响。方法:选用MDA-MB-231细胞株,分别通过不同浓度的Zebularine作用细胞株24 h后收集细胞,运用MTT法测定Zebularine对乳腺癌细胞株MDA-MB-231细胞的增殖;通过Western Blot检测Zebularine对乳腺癌细胞株SARI的表达。结果:随着药物浓度的增加,Zebularine能够明显抑制乳腺癌细胞株MDA-MB-231的增殖(P<0.05),Zebularine能够显著提高乳腺癌细胞株MDA-MB-231 SARI蛋白的表达(P<0.05)。结论:Zebularine通过抑制乳腺癌细胞增殖,促进SARI蛋白的表达,从而发挥肿瘤抑制作用。Objective:To investigate the effect of Zebularine on the proliferation and SARI expression of human breast cancer cell line MDA-MB-231.Method:MDA-MB-231 cell line were cultured in vitro and treated with different doses Zebularine for 24 hours.MTT method was used to detect the cell proliferation,and western blot was used to detect the expression of SARI in human breast cancer cell line MDA-MB-231.Result:Different doses Zebularine could significantly inhibit the proliferation of human breast cancer cell line MDA-MB-231 with the increase of drug concentration(P<0.05).Zebularine could obviously reduced the expression of SARI in human breast cancer cell line MDA-MB-231(P<0.05).Conclusion:Zebularine might inhibit the proliferation, improve the expression of SARI,then inhibit the malignant proliferation of tumor cells,then exerting tumor inhibitory effect.
