摘  要:目的采用高效液相色谱-高分辨飞行时间质谱(HPLC-Q-TOF MS)对黄花败酱总皂苷部位中的化学成分进行快速鉴别。方法采用ACQUITY UPLCTM BEH C_(18)(2.1 mm×50 mm,1.7μm)色谱柱,以乙腈-(0.1%甲酸,10 mmol甲酸铵)水溶液为流动相梯度洗脱,柱温:35℃,进样量:3μL,流速:0.3 mL/min,采用ESI源在正离子模式下采集数据。根据化合物准确的相对分子质量及二级碎片离子信息,参考文献数据及与对照品对照,鉴定出黄花败酱总皂苷部位的主要化学成分。结果共鉴定出27个化学成分,其中三萜皂苷类成分10种,甾体皂苷2种,甾醇类3种,单萜类4种,有机酸类2种,糖苷类2种,黄酮类1种,其他类3种。结论 HPLC-Q-TOF MS技术为鉴定黄花败酱有效部位化学成分提供了一种快速、高效的方法。Objective To identify the chemical components of total saponins from Patrinia scabiosaefolia Fisch by high performance liquid chromatography-time of flight mass spectrometry (HPLC-TOF/MS).Methods The separation was performed on ACQUITY UPLCTM BEH C 18 column(2.1 mm×50 mm, 1.7 μm),and the gradient elution of acetonitrile-(0.1% formic acid,10 mmol ammonium formate) was used as mobile phase.The column temperature was 35 ℃,the flow rate was 0.3 mL/min and the injection volume was 3 μL.The data were collected by the positive and ion mode by using Q-TOF/MS and ESI source.The main chemical constituents of total saponins of Patrinia scabiosaefolia Fisch were identified according to the exact molecular mass,the cleavage fragments of MS/MS,the literature data and the reference control.Results Twenty seven chemical components were identified under the optimized liquid-mass spectrometry conditions,which were ten kinds of triterpenoid saponins,two kinds of steroidal saponins,three kinds of sterols,four kinds of monoterpenes,two kinds of organic acids,two kinds of glycosides,one kind of flavonoids and three kinds of other types.Conclusion The established HPLC-Q-TOF MS method provides a fast and efficient method for identifying the chemical constituents of the active part of the Patrinia scabiosaefolia Fisch.
