摘 要:目的结直肠癌组织中let-7表达明显降低,具有类似于抑癌基因作用。let-7启动子区多态性与多种肿瘤易感性有关,本研究旨在探讨let-7启动子区多态性与结直肠癌的相关性。方法收集2012-03-01-2016-08-31云南省肿瘤医院结直肠外科收治的154例结直肠癌(结肠癌51例,直肠癌103例)患者外周静脉血,收集203例同期健康体检人群外周静脉血作为对照。应用TaqMan探针法对let-7启动子区多态性rs10877887和rs13293512进行分型,统计分析其与结直肠癌发病的相关性。结果rs10877887位点TT、CT、CC基因型、T和C等位基因在病例组中的频率分别为37.0%、42.2%、20.8%、58.1%和41.9%,在对照组中的频率分别为49.8%、39.9%、10.3%、69.7%和30.3%。CC与TT相比,校正OR=2.55,95%CI为1.33~4.90;CT/CC与TT相比,校正OR=1.69,95%CI为1.10~2.60;C与T相比,校正OR=1.64,95%CI为1.20~2.24;rs10877887CC、CT/CC基因型和C等位基因增加了结直肠癌的发病风险。未发现rs13293512多态性与结直肠癌发病之间存在明显关联。结论 let-7启动子区rs10877887CC或者CT/CC基因型可能是结直肠癌的易感因素之一。OBJECTIVE The let-7 level is downregulated in colorectal cancer(CRC),functioning as a tumor suppressor.Genetic polymorphisms in the promoter of let-7 were reported to be related to the risk of several tumors.The aim of this study was to investigate the association between polymorphisms in the promoter of let-7 and risk of CRC.METHODS The rs10877887 and rs13293512 polymorphisms were genotyped using TaqMan assay.The association between the rs10877887 and rs13293512 polymorphisms and CRC risk was analyzed by SPSS software.RESULTS The frequencies of the rs10877887 TT,CT,CC,T and C were 37.0%,42.2%,20.8%,58.1% and 41.9% in CRC patients and 49.8%,39.9%,10.3%,69.7% and 30.3%in controls.The rs10877887 CC,CT/CC genotypes and C allele were associated with increased risks of CRC(CCvs TT:adjusted OR=2.55,95%CI:1.33-4.90;CT/CCvs TT:adjusted OR=1.69,95%CI:1.10-2.60;C vs T:adjusted OR=1.64,95%CI:1.20-2.24).No significant association between the rs13293512 polymorphism and risk of CRC was observed.CONCLUSION The rs10877887 CC or CT/TT genotypes in the promoter of let-7 may be a risk factor for the development of CRC.
