摘  要:目的我国是世界上食管癌发病率和死亡率最高的国家,且四川川北地区和盐亭县为我国食管癌高发区之一。本研究检测端粒保护蛋白TPP1在食管癌组织中的表达,分析其与临床病理特征和食管癌肿瘤标志物的相互关系,探讨其临床意义。方法应用RT-PCR和蛋白质印迹法分别检测端粒保护蛋白TPP1在65例食管癌组织及其配对癌旁组织中mRNA和蛋白表达,化学发光免疫分析仪定量检测食管癌外周血相关肿瘤标志物CEA、SCC、CYRFA21-1和TPA等的浓度。结果食管癌组织TPP1的mRNA表达水平为1.98±1.35,明显高于癌旁组织的1.07±0.44,t=5.322,P<0.001。TPP1在癌组织的相对蛋白表达水平为1.31±1.24,高于癌旁组织的1.00±0.87,t=3.896,P<0.01。TPP1高表达与患者年龄负相关,与肿瘤淋巴结转移正相关。进一步分析发现,食管癌组织中TPP1的高表达水平与外周血血清中CEA、CYFRA21-1和TPA浓度有一定相关性。结论 TPP1可能参与了食管癌的发生发展,有望成为食管癌诊治潜在的分子靶标。OBJECTIVE China has the highest incidence and mortality of esophageal cancer in the world,and North Sichuan and Yanting County are the high incidence areas of esophageal cancer in China.This study aimed to investigate the telomere binding protein TPP1 expression in esophageal carcinoma tissue,and analyze the correlation of TPP1 with the clinico-pathological characteristics and the relationship with serum tumor markers,further to explore the potential clinical significance of TPP1 in esophageal carcinoma.METHODS RT-PCR and Western blot were used to detect the TPP1 mRNA and protein expression in pairs of primary esophageal carcinoma and adjacent tissue specimens respectively.Esophageal carcinoma related serum tumor markers such as CEA,SCC,CYRFA21-1 and TPA were quantitatively measured with chemiluminescent analyzer.RESULTS The expression level of telomere protective protein TPP1 in esophageal cancer tissues was 1.98±1.35,which was significantly higher than that in adjacent tissues(t=5.322,P<0.001).The relative expression level of TPP1 in cancer tissues was 1.31±1.24,which was higher than that in adjacent tissues(t=3.896,P<0.01).Upregulation of TPP1 significantly correlated with patients’ age and lymph nodes metastasis.The higher expression of TPP1 in esophageal cancer tissues was correlated with the concentration of tumor marker CEA,CYFRA21-1 and TPA in peripheral blood serum.CONCLUSION Telomere binding protein TPP1 may play an important role in the tumorigenesis and development of esophageal carcinoma,targeting TPP1 would be potential for the diagnosis and treatment of esophageal carcinoma.
