直孔藏纸抑菌性及毒理学安全性评价

摘　　要：采用顶空气相色谱技术,检测以瑞香狼毒根部韧皮为原料所造直孔藏纸的抑菌效果,评价其抑菌性;通过家兔皮肤刺激试验、豚鼠皮肤变态反应试验及二者受试区皮肤的病理组织学变化,综合评价直孔藏纸毒理学安全性。研究结果表明,5 mL的2.9×10^(2 )CFU/mL大肠埃希氏菌菌液在培养容器为顶空瓶(20 mL)的条件下,通过气相色谱仪检测二氧化碳信号,简便计算出直孔藏纸抑菌率42.07±0.026%、雪梨纸16.14±0.021%、宣纸12.09±0.046%和新闻纸8.19±0.021%,直孔藏纸抑菌率同等条件下显著高于其他3种纸张;直孔藏纸对家兔皮肤的最高刺激指数为0、无刺激性反应,对豚鼠的皮肤变态反应试验致敏率为0%、致敏强度极轻,且直孔藏纸受试区皮肤病理组织学无异常变化。基于以上研究结果,认为直孔藏纸具有明显抑菌性和一定的安全性。The bacteriostasis effect of Zhikong Tibetan paper made from handmade paper from the root phloem of Stellera chamaejasme was determined by headspace gas chromatography(HS-GC);Comprehensive evaluation of toxicologiceadsal safety of Zhikong Tibetan paper by rabbit skin irritation test, guinea pig skin allergy test and pathological histological changes of skin in the test area. 5 mL of 2.9×10^2 CFU/mL Escherichia coli in a culture vessel with a hpace vial(20 mL). The gas chromatographic signal was used to detect the carbon dioxide signal, and the inhibition rate of the Zhikong Tibetan paper with obvious bacteriostasis was easily calculated 42.07±0.026%, which was higher than the Tissue paper 16.14±0.021%, Rice paper 12.09±0.046%, and Newsprint 8.19±0.021% under the same conditions. The highest stimulation index of Zhikong Tibetan paper on the skin of rabbits was 0. There is no irritating reaction. The allergic reaction test rate of guinea pigs skin is 0%. The sensitization intensity is extremely light. And there is no abnormal change in the pathological histology of the test area of Zhikong Tibetan paper. Based on the above research, it is believed that the Zhikong Tibetan paper has obvious bacteriostasis and certain safety.
